白蓼花对小鼠银屑病的治疗作用及机制实验研究

Abstract

Objective:To investigate the chemical components of Koenigia coriaria(Grig.)T.M.Schust.& Re-veal and its therapeutic effect on psoriasis in mice.Methods:Extracts of Koenigia coriaria(Grig.)T.M.Schust.&Reveal were prepared.UPLC-MS/MS combined with positive and negative ion detection modes was used to identify the chemical components in the extracts under specific chromatographic and mass spectrometric conditions.Forty male mice were selected and randomly divided into the blank control group,model group,positive drug group,Koeni-gia coriaria(Grig.)T.M.Schust.& Reveal medium-dose group and Koenigia coriaria(Grig.)T.M.Schust.& Re-veal high-dose group,with 8 mice in each group.Except for the blank control group,mice in other groups were in-duced to establish psoriasis models by smearing imiquimod cream on the back;mice in the blank control group were smeared with an equal amount of vaseline on the back daily.All groups received intragastric administration once a day for 6 consecutive days before smearing.The positive drug group was given 1 mg/kg methotrexate tablet solution in-tragastrically;the medium-dose and high-dose groups were given 2.25 g/kg and 4.5 g/kg Koenigia coriaria(Grig.)T.M.Schust.& Reveal extract solution intragastrically,respectively;the blank control group and model group were given an equal volume of 0.9%sodium chloride solution intragastrically.Daily changes in skin lesions of mice were observed,and the Psoriasis Area and Severity Index(PASI)score was evaluated.HE staining was used to observe histopathological changes of mouse skin and measure epidermal thickness.Immunofluorescence staining was used to detect the expression of Ki-67 in skin lesion tissues.Results:A total of 32 components were identified in the crude ex-tract of Koenigia coriaria(Grig.)T.M.Schust.& Reveal,mainly including flavonoids and their derivatives(e.g.,quercetin,rutin,kaempferol and their glycosides),as well as phenolic acids,nucleosides,amino acids,terpenes and or-ganic acids.In the mouse experiment,compared with the model group,the PASI scores of mice in the Koenigia cori-aria(Grig.)T.M.Schust.& Reveal medium-dose group,high-dose group,and the positive drug group were de-creased;the epidermal thickness was reduced;the phenomena of disordered arrangement of epidermal cells,hyperker-atosis and parakeratosis were improved;dermal vascular dilation was alleviated;inflammatory cell infiltration was sig-nificantly reduced;and the expression level of Ki-67 in the lesion tissues was decreased(all P<0.05).Among the posi-tive drug group,Koenigia coriaria(Grig.)T.M.Schust.& Reveal medium-dose group and Koenigia coriaria(Grig.)T.M.Schust.& Reveal high-dose group,there were no statistically significant differences in PASI scores,epidermal thick-ness,and expression level of Ki-67 in lesion tissues in comparison(all P>0.05).Conclusion:The crude extract of Koe-nigia coriaria(Grig.)T.M.Schust.& Reveal contains multiple components,and flavonoids and phenolic acids are the main active substances.Koenigia coriaria(Grig.)T.M.Schust.& Reveal can inhibit keratinocyte proliferation,reduce epidermal thickening,and improve inflammatory responses,thereby alleviating skin lesion symptoms in psoriatic mice.
