穿心莲内酯对子宫肌瘤细胞增殖、迁移和侵袭的影响及机制实验研究
Effects and mechanisms of andrographolide on proliferation,migration and invasion of uterine fibroid cells

Abstract
Objective:To investigate the effects of andrographolide on the proliferation,migration and invasion of uterine fibroid cells and its possible mechanisms.Methods:Uterine fibroid tissue samples were collected from patients with uterine fibroids after surgery,and primary uterine fibroid cells were extracted.Primary uterine fibroid cells were treated with different concentrations of andrographolide(0,10,20,40,80 μmol/L)for 24 hours.Cell viability was de-tected at different concentrations,and the half-maximal inhibitory concentration(IC50)of andrographolide on primary uterine fibroid cell viability was calculated.Both primary uterine fibroid cells and uterine fibroid cell lines were ran-domly divided into control,low-concentration and high-concentration groups.The control group was cultured routine-ly,the low-concentration group was treated with 5 μmol/L andrographolide during routine culture,and the high-con-centration group was treated with 10 μmol/L andrographolide.Cell viability was detected using a cell counting kit.Cell migration and invasion abilities were evaluated by Trans well assay and Matrigel assay,respectively.The expres-sion of proteins related to the nuclear factor-KB(NF-κB)/cyclooxygenase-2(COX-2)and phosphatidylinositol-3 ki-nase(PI3K)/protein kinase B(AKT)signaling pathways in cells was detected by Western blot.Results:After 24 hours of treatment with different concentrations(0,10,20,40,80 μmol/L)of andrographolide,the IC50 for prima-ry uterine fibroid cells was 6.833 μmol/L.In both primary uterine fibroid cells and uterine fibroid cell lines,the via-bility of uterine fibroid cells,the number of migrating cells,the number of invading cells,and the expression levels of NF-κB-p65,COX-2,PI3K-p85,and p-AKT proteins were decreased in the low-and high-concentration groups com-pared with the control group,while the expression level of phosphatase and tensin homolog(PTEN)protein was in-creased(all P<0.05).The changes in the above indicators were more significant in the high-concentration group than in the low-concentration group(all P<0.05).Conclusion:Andrographolide may inhibit the proliferation,migra-tion and invasion of uterine fibroid cells by regulating the NF-κB/COX-2 and PI3K/AKT signaling pathways.关键词
