Nedaplatin inhibits the proliferation,migration and invasion of melanoma cells by activating oxidative stress and Caspase-9-mediated mitochondrial apoptosis

Abstract
Objective:To investigate the effects of nedaplatin on melanoma cells through the activation of oxida-tive stress and Caspase-9-mediated mitochondrial apoptosis.Methods:Melanoma A375 cells were randomly divided into five groups:control group,nedaplatin group,nedaplatin+lipoic acid group,nedaplatin+si-NC group and nedapla-tin+si-Caspase-9 group.The control group received no drug treatment.The nedaplatin group was treated with 10 μmol/L nedaplatin for 24 h.The nedaplatin+lipoic acid group was treated with 10 μmol/L nedaplatin combined with 50 μmol/L lipoic acid for 24 h.The nedaplatin+si-NC group was treated with 10 μmol/L nedaplatin and 100 nmol/L si-NC for 24 h.The nedaplatin+si-Caspase-9 group was treated with 10 μmol/L nedaplatin and 100 nmol/L si-Caspase-9 for 24 h.Oxidative stress markers(reactive oxygen species[ROS],malondialdehyde[MDA],superox-ide dismutase[SOD]),mitochondrial apoptosis-related proteins(Caspase-9,cleaved Caspase-3,Bcl-2,Bax),mito-chondrial membrane potential,ATP content,cell proliferation activity,migration rate and invasion rate were measured in each group.Results:Compared with the control group,the nedaplatin group showed increased levels of ROS and MDA,increased protein expression levels of Caspase-9 and cleaved Caspase-3,increased Bax/Bcl-2 ratio,and de-creased levels of SOD,mitochondrial membrane potential,ATP content,cell proliferation activity,cell migration rate and cell invasion rate(all P<0.05).Compared with the nedaplatin group,the nedaplatin+lipoic acid group showed increased levels of SOD,cell proliferation activity,cell migration rate,and cell invasion rate,and decreased levels of ROS and MDA(all P<0.05).Compared with the nedaplatin+si-NC group,the nedaplatin+si-Caspase-9 group showed decreased protein expression levels of Caspase-9 and cleaved Caspase-3,decreased Bax/Bcl-2 ratio,and in-creased mitochondrial membrane potential,ATP content,cell proliferation activity,cell migration rate and cell inva-sion rate(all P<0.05).Conclusion:Nedaplatin inhibits the proliferation,migration and invasion of melanoma cells by activating oxidative stress and Caspase-9-mediated mitochondrial apoptosis.关键词
