宝藿苷Ⅱ对口腔鳞状细胞癌细胞增殖、迁移和凋亡的影响及机制实验研究
Effects and mechanisms of baohuoside Ⅱ on proliferation,migration and apoptosis of oral squamous cell carcinoma cells

Abstract
Objective:To investigate the effects and possible mechanisms of baohuoside Ⅱ on the proliferation,migration and apoptosis of oral squamous cell carcinoma(OSCC)cells.Methods:CAL27 cells were treated with dif-ferent concentrations(0,1,2,5,10,25,50 μmol/L)of baohuoside Ⅱ for 24 and 48 hours.Cell viability and the half-maximal inhibitory concentration(IC50)were detected by the CCK-8 method.CAL27 cells were treated with different concentrations(0,5,10 μmol/L)of baohuoside Ⅱ for 24 and 48 hours,and cell migration ability was detected by scratch assay.CAL27 cells were randomly divided into negative control(NC)group,low concentration baohuoside Ⅱ(BHE-L)group,medium concentration baohuoside Ⅱ(BHE-M)group,and high concentration baohuoside Ⅱ(BHE-H)group.The NC group undergoes routine cultivation.According to the 48-hour IC50 value of baohuoside Ⅱ,the BHE-L,BHE-M and BHE-H groups were cultured with media containing 5,10 and 25 μmol/L baohuoside Ⅱ,re-spectively.Apoptosis of cells in each group was detected by flow cytometry.The mRNA expression of β-catenin,c-myc and cyclin D1 was detected by real-time quantitative PCR(RT-qPCR),and the protein expression levels ofβ-catenin,c-myc and cyclin D1 were detected by Western blot.Results:Compared with 0 μmol/L baohuoside Ⅱ,the viability of CAL27 cells treated with 2,5,10,25,and 50 μmol/L baohuoside Ⅱ(for 24 and 48 hours)was decreased(all P<0.05).Moreover,at the same concentration,the viability of CAL27 cells treated with baohuoside Ⅱ for 48 hours was lower(all P<0.05).The IC5o of baohuoside Ⅱ for CAL27 cells was 16.89 μmol/L at 24 hours and 9.71 μmol/L at 48 hours.Compared with 0 μmol/L baohuoside Ⅱ,the migration rate of CAL27 cells treated with 5 and 10 μmol/L baohuoside Ⅱ(for 24 and 48 hours)was decreased(all P<0.05).Moreover,at the same concen-tration,the migration rate of CAL27 cells treated with baohuoside Ⅱ for 48 hours was lower(all P<0.05).Com-pared with the NC group,the apoptosis rate of CAL27 cells in the BHE-L,BHE-M and BHE-H groups increased successively,and the mRNA and protein expression levels of β-catenin,c-myc and cyclin D1 decreased successively(all P<0.05).Conclusion:baohuoside Ⅱ may inhibit the proliferation and migration of OSCC cells and induce apop-tosis by downregulating the Wnt/β-catenin signaling pathway.关键词
