Abstract

Objective:To investigate the effects of sevoflurane exposure at different stages of pregnancy on off-spring rats and its possible mechanisms.Methods:A total of 64 SPF-grade SD rats,with an equal number of males and females,were selected.The male and female rats were housed together to promote mating and successful preg-nancy.The pregnant female rats were randomly divided into four groups:the control group,the early pregnancy group(6 d),the mid-pregnancy group(12 d),and the late-pregnancy group(18 d),with 8 rats in each group.The control group did not inhale sevoflurane.The other groups were placed in an anesthesia chamber and exposed to 3%sevoflurane at different stages,for 6 hours each time,once a day,for a total of 5 days.On day 31 after birth,24 pups were randomly selected from each group to undergo the water maze test,and serum immune function indicators were measured.Hippocampal tissues were taken from the pups in each group for HE staining and Nissl staining to observe the structure.Golgi staining was used to detect dendritic spine density in the hippocampal tissue.Western blot was used to detect the expression of BDNF and TrkB proteins in the hippocampal tissue.Results:Compared with the con-trol group,the escape latency was prolonged,and the time to first reach the platform and the time spent in the plat-form quadrant were increased,while the number of platform crossings was decreased in the early and mid-pregnancy groups(all P<0.05).Compared with the control group,the level of CD3+was increased,while the levels of CD4+and the CD4+/CD8+ratio were decreased in the early and mid-pregnancy groups(all P<0.05).Compared with the control group,the dendritic spine density in the hippocampal tissue of the early and mid-pregnancy groups was re-duced(all P<0.05).Compared with the control group,the expression levels of BDNF and TrkB proteins in the hipp-ocampal tissue of the early and mid-pregnancy groups were decreased(all P<0.05).There were no significant differences in the above indicators between the control group and the late-pregnancy group(all P>0.05).The hipp-ocampal tissue structure of the control group pups was normal,while the hippocampal tissue structure of the late-pregnancy group pups showed slight damage.The hippocampal tissue of the early and mid-pregnancy groups was se-verely damaged,with degenerative and necrotic neurons and a large number of inflammatory cell infiltrations.The control group and late-pregnancy group pups had large areas of blue Nissl bodies in the cytoplasm of neurons in the hippocampal tissue,while the number of Nissl bodies in the early and mid-pregnancy groups gradually decreased and were loosely arranged.Conclusion:Sevoflurane exposure during pregnancy has stage-specific effects on offspring rats.Exposure during early and mid-pregnancy may damage hippocampal neurons,inhibit dendritic spine development,and affect the immune and cognitive functions of offspring rats.The mechanism may be related to the downregulation of the BDNF/TrkB signaling pathway.
