牛膝叶提取物对异丙肾上腺素诱导大鼠心肌梗死纤维化的影响

Abstract
Objective:To observe the effect of Achyranthes bidentata leaf extract on myocardial infarction fibrosis induced by isoproterenol(ISO)in rats.Methods:Twenty-four SD rats were randomly divided into model group,control group,Achyranthes bidentata leaf extract group and captopril group,with 6 rats in each group.The rats in the model group,the Achyranthes bidentata leaf extract group and the captopril group were modeled with isoproterenol,and the control group was subcutaneously injected with an equal volume of normal saline.After successful modeling,the Achyranthes bidentata leaf extract group and captopril group were given corresponding drugs by gavage,and the remaining two groups were given normal saline by gavage.The histological changes of myocardium were observed by HE and Masson staining.The indexes of serum myocardial injury,oxidative stress and inflammatory factors in myocardial tissue were determined by ELISA.WB was used to detect the expression of TGF-β 1,p-Samd3,collagen Ⅰ and α-SMA.Results:In the control group,the morphology of myocardial cells was normal and arranged neatly.In the model group,the myocardial cells were swollen,the gap was enlarged,and the myocardial fibers were broken with cell necrosis.The myocardial injury in the Achyranthes bidentata leaf extract group and captopril group was significantly reduced.There was no myocardial fibrosis in the control group,while the model group showed significant myocardial fibrosis,cardiomyocyte hypertrophy and myofibril destruction.The degree of myocardial fibrosis in the Achyranthes bidentata leaves extract group and captopril group was significantly reduced.The levels of CK,LDH,CK-MB and cTnT in the model group were higher than those in the control group(P<0.05).The levels of CK,LDH,CK-MB and cTnT in the Achyranthes bidentata leaves extract group and captopril group were lower than those in the model group(P<0.05).Compared with the control group,the levels of SOD,CAT and GSH-PX in the model group were decreased(P<0.05),and the levels of TNF-α,MAD and IL-6 were increased(P<0.05).Compared with the model group,the levels of SOD,CAT and GSH-PX in the captopril group and the Achyranthes bidentata leaf extract group were increased(P<0.05),and the levels of MAD,TNF-α and IL-6 were decreased(P<0.05).The expression of TGF-β 1,α-SMA,collagen Ⅰ and Smad3 protein in myocardial tissue of rats in the model group was higher than that in the control group(P<0.05).The expression of TGF-β 1,α-SMA,collagen Ⅰ and Smad3 protein in myocardial tissue of Achyranthes bidentata leaf extract group and captopril group was lower than that in model group(P<0.05).Conclusion:Achyranthes bidentata leaf extract could inhibit myocardial fibrosis by inhibiting inflammatory response and oxidative stress injury.关键词
