肾衰营养胶囊调节炎症及肌分化改善慢性肾脏病大鼠骨骼肌萎缩的机制研究

Abstract
Objective To investigate the mechanism by which Shenshuai Yingyang Capsule(肾衰营养胶囊,SSYYC)alleviates skel-etal muscle atrophy in rats with chronic kidney disease(CKD).Methods The CKD rat model was established by 5/6 nephrectomy.Suc-cessfully modeled rats were randomly divided into a model group and a SSNC group,with 8 rats in each group;a group(n=8)was estab-lished.After 10 weeks of treatment,serum creatinine(Scr),urea nitrogen(BUN),serum albumin(Alb),and inflammatory marker levels(IL-1β,IL-6,CRP,TNF-α)were measured in each group.The skeletal muscle morphology of rats in each group was observed using HE staining.Target genes were identified using transcriptome sequencing,and differentiation protein levels were detected by Western blot(WB).Results Compared with the sham surgery group,after 8 weeks,the model group showed significantly decreased renal function,upregulated serum inflammatory indicators(IL-1β,IL-6,CRP,TNF-α),reduced cross-sectional area of skeletal muscle fibers,and downregulated MyoD and Myf4 protein levels.Compared with the model group,the SSYYC intervention group showed improved renal function and decreased levels of inflammatory factors;the cross-sectional area of skeletal muscle fibers increased(P<0.05),and MyoD and Myf4 protein levels were upregulated(P<0.05).Transcriptome sequencing results showed 201 differen-tially expressed genes(DEGs)in skeletal muscle tissue between the model group and the SSYYC group.Among these,44 were target genes,including Fbxo32 and Tmco3.Gene enrichment analysis suggested an association with pathways regulating skeletal muscle myo-genesis,differentiation,and metabolism.Conclusion SSYYC alleviates skeletal muscle inflammation and atrophy in CKD rats,which may be related to the regulation of MyoD and Myf4.关键词
