醒脑解郁方对PSD模型大鼠皮层及海马神经元突触可塑性的影响
Effects of Xingnao Jieyu Decoction(醒脑解郁方)on synaptic plasticity in cortex and hippo-campal neurons of post-stroke depression model rats

Abstract
Objective To observe the effects of Xingnao Jieyu Decoction(醒脑解郁方,XNJY)on depression-like behaviors in post-stroke depression(PSD)model rats and on synaptic plasticity in cortical and hippocampal neurons.Methods Seventy-two SD rats were randomly assigned to the blank group(0.9%NaCl),ischemic stroke group(0.9%NaCl),model group(PSD model,0.9%NaCl),fluoxetine group(2.08 mg/kg),XNJY low-dose(XNJY-L)group(10.5 g crude herb/kg),and XNJY high-dose(XNJY-H)group(20.1 g crude herb/kg).Except the blank group,all other groups underwent middle cerebral artery occlusion to establish the cerebral ischemia-reperfusion(CIR)model.Except the blank group and ischemic stroke group,the remaining groups were subjected to chronic unpredictable mild stress to establish the PSD model.After 21 days of drug treatment,rat body weight and behavioral assessments were performed at baseline(before modeling),day 10,and day 21.Nissl staining was employed to observe morphological changes in Nissl bodies within the cortex and hippocampus.Western blot(WB)was performed to detect the expression levels of synaptophysin(SYN)and postsynaptic density protein 95(PSD-95)in the cortex and hippocampus.Results Compared with the model(PSD)group,rats in the fluoxetine(Flu),XNJY-L/H groups showed increased body weight(P<0.05,P<0.01).In behavioral assessments,compared with the model group,the immobility time in the forced swim test was significantly reduced in the Flu,XNJY-L/H groups(P<0.05,P<0.01).Sucrose preference test(SPT)consumption was significantly increased in the Flu and XNJY-L groups(P<0.01),while in the open field test(OFT),both horizontal and vertical movement scores were significantly increased in the Flu,XNJY-L/H groups(P<0.05,P<0.01).Nissl staining results showed that neurons in the cortex and hippocampus of rats in the Flu and XNJY-L groups were arranged in an orderly manner and exhibited more abundant Nissl bodies.WB analysis indicated that,compared with the model group,the Flu and XNJY-L groups exhibited upregulated expression levels of SYN and PSD-95 proteins in the cortex and hippocampus(P<0.05,P<0.01).Conclusion XNJY ameliorates depression-like behaviors in PSD model rats,possibly by improving synaptic plasticity via upregulation of SYN and PSD-95 expression in the cortex and hippocampus.关键词
