桃核承气汤干预深静脉血栓形成的作用及机制研究
Effect and mechanism of Taohe Chengqi Decoction(桃核承气汤)in intervening deep vein thrombosis

Abstract
Objective To evaluate the preliminary mechanism of Taohe Chengqi Decoction(桃核承气汤,THCQD)in intervening deep vein thrombosis(DVT).Methods High-performance liquid chromatography(HPLC)was used to analyze the chemical components of THCQD.In the mice coagulation experiment,a normal group,low/high-dose groups of THCQD,and a rivaroxaban group were estab-lished.One hour after a single dose administration,capillary blood was collected from the orbit,and the coagulation time of each group was measured.In the DVT experiment,a sham surgery group,model group,low/high-dose groups of THCQD,and rivaroxaban group were established.Continuous administration was given for 3 days,with modeling performed on day 2.One hour after the final dose,thrombus weight and thrombus length were measured.Hematoxylin and eosin(HE)staining was used to observe pathological changes in the venous wall(including thrombus),and Masson's staining was used to observe collagen fiber deposition within the thrombus.Finally,network pharmacology was applied to explore the mechanism of THCQD in inhibiting DVT,and experimental validation of its key target proteins was performed.Results THCQD was identified to contain 15 compounds,including gallic acid,rhein,and emodin.Experimental results showed that THCQD could prolong the coagulation time in normal mice,reduce thrombus weight and length in DVT rats,inhibit inflammatory cell infiltration at the lesion site,and decrease collagen fiber deposition.Network pharmacology analysis iden-tified key targets for the intervention of THCQD in DVT,including protein kinase B(AKT),signal transducer and activator of transcrip-tion 3(STAT3),and transcription factor AP-1(JUN).Experimental validation further demonstrated that THCQD could up-regulate the expression of phosphorylated AKT(p-AKT)protein in the lesion area of DVT rats.Conclusion THCQD exerts anti-DVT effect by up-regulating the expression of p-AKT protein and then inhibiting inflammation.关键词
