Abstract

Objective To compare effects of Sijunzi Decoction(四君子汤,SD)and its different components based on the medicated serum of spleen deficiency mice and cell models.Methods An IEC-6 cell scratch model and a corticosterone-induced PC12 cell injury model were established.Blank and model group cells were treated with blank or spleen-deficient mouse serum,while the treatment groups were administered drug-containing serum from spleen-deficiency mice treated with SD,non-polysaccharide fraction of SD(NPS),or active fraction of SD(ECS).The migration of IEC-6 cells was recorded,and Western blot(WB)was used to detect the pro-tein expression levels of the PI3K/AKT signaling pathway in IEC-6 cells.PC12 cell viability was measured using the Cell Counting Kit-8(CCK-8)assay,and lactate dehydrogenase(LDH)release rate was determined by ELISA.Results Compared with the model group of IEC-6 cells,IEC-6 cells in SD group,NPS group and ECS group showed significant migration phenomenon(P<0.05),and NPS medi-cated serum showed better effect than SD and ECS on promoting the migration.All three medicated serum could activate PI3K/AKT sig-naling pathway of IEC-6 cells(P<0.05)to repairing the intestinal epithelial cell injury.Compared with the model group of PC12 cells,SD,NPS and ECS medicated serum significantly improved corticosterone-induced PC12 cell injury(P<0.05)and reduced LDH release rate of PC12 cells(P<0.05).Conclusion Drug-containing serum from spleen-deficiency mice treated with SD and its different compo-nents exerts protective effects on IEC-6 and PC12 cell injury.The efficacy of ECS is comparable to that of SD,suggesting it may be the active fraction.The PI3K/AKT pathway may be one of the mechanisms by which ECS repairs intestinal epithelial cells.
