大建中汤方中有无饴糖对腹泻型肠易激综合征大鼠的作用机制研究
Therapeutic effect of Dajianzhong Decoction(大建中汤方)with or without maltose on diarrhea-predominant irritable bowel syndrome in rats and its mechanism

Abstract
Objective To investigate the therapeutic effect of Dajianzhong Decoction(大建中汤方,DJZD)with or without maltose on rats with diarrhea-predominant irritable bowel syndrome(IBS-D)presenting spleen yang deficiency syndrome and explore its mecha-nism.Methods An IBS-D rat model with spleen yang deficiency syndrome was established.After interventions with DJZD containing maltose(full formula)or DJZD without maltose,fecal characteristics,abdominal withdrawal reflex(AWR)scores,organ indices,and colonic histopathology were assessed.Gut microbiota composition was analyzed using metagenomic sequencing.Flow cytometry(FCW)quantified Th17 and Treg cell proportions in colon tissue.mRNA expressions of IL-6,IL-17,IL-10,TGF-β,and RORγt were mea-sured by RT-qPCR.Results Compared to the normal group,the model group showed significantly increased Bristol stool scores,fecal wet weight,and AWR scores.Both DJZD treatments reduced these parameters,with the full formula exhibiting greater efficacy.At the phylum level,both treatments decreased the Bacillota/Bacteroidota ratio.At the genus level,Bacteroides and Ruminococcus were increased,while Lactobacillus was decreased.Both treatments reduced Th17 cell proportions,elevated Treg cell proportions,lowered the Th17/Treg ratio,and downregulated IL-6,IL-17,IL-10,TGF-β,and RORγt mRNA expressions versus the model group.Conclu-sion Both DJZD with and without maltose alleviated IBS-D in rats with spleen yang deficiency.The mechanism may be associated with modulation of gut microbiota composition and restoration of colonic Th17/Treg balance.关键词
