温阳通便汤调控PINK1/Parkin通路治疗功能性便秘的机制研究
Mechanism of Wenyang Tongbian Decoction(温阳通便汤)in alleviating functional constipa-tion via regulating the PINK1/Parkin pathway

Abstract
Objective To investigate the mechanism by which Wenyang Tongbian Decoction(温阳通便汤,WYTBD)alleviates func-tional constipation(FC)through regulating the PINK1/Parkin signaling pathway to inhibit apoptosis.Methods C57BL/6 mice were ran-domly divided into the control group,model group,low/medium/high-dose WYTBD groups,and the lactulose group.Except for the con-trol group,all other groups were administered loperamide hydrochloride by gavage once daily for one week to establish the FC model.Following successful model establishment,the mice were treated with graded doses of WYTBD or lactulose for one week.Subsequently,the number of fecal pellets,fecal water content,and intestinal transit rate were measured.HE staining was used to observe pathological changes in colon tissue,TUNEL staining was used to detect colonocyte apoptosis,and Western blot(WB)was performed to measure the expression of PINK1,Parkin,Bcl-2,Bax,and Caspase-3 proteins in colon tissue.Results WYTBD significantly increased the number of fecal pellets,fecal water content,and intestinal transit rate in constipated mice(P<0.05 or P<0.01),ameliorated pathological changes in colon tissue,inhibited colonocyte apoptosis,upregulated the expression of PINK1,Parkin,and Bcl-2 proteins(P<0.05),and downregulated the expression of Bax and Caspase-3 proteins(P<0.05).Conclusion WYTBD alleviates FC by regulating the PINK1/Parkin pathway to inhibit apoptosis.关键词
