Effects of curcumin on proliferation,apoptosis,and angiogenesis of lung cancer cells via regu-lating the HIF-1α/VEGFR/Akt pathway

Abstract
Objective To investigate the effects of curcumin on proliferation,apoptosis,angiogenesis,and the hypoxia-inducible factor-1α/vascular endothelial growth factor receptor/protein kinase B(HIF-1α/VEGFR/Akt)pathway in lung cancer cells.Methods A549 lung cancer cells were cultured and divided into the following groups:the control group(Control),curcumin low/medium/high-concentration treatment groups(CUR-L/M/H),and a curcumin high-concentration treatment combined with pathway activator dimethyl-oxalylglycine group(CUR-H+DMOG).Cell proliferation was detected using the Edu assay;cell apoptosis was measured by flow cytom-etry(FCM);angiogenesis was observed;and the expression of vascular endothelial growth factor A(VEGFA)and proteins related to the HIF-1α/VEGFR/Akt pathway was determined by Western blot(WB).Results Compared with the Control group,the CUR-L/M/H groups showed reduced angiogenic lumen formation,decreased Edu-positive rates,and lower expression levels of CyclinD1,Bcl-2,VEGFA,HIF-1α,VEGFR2,and p-AKT/AKT,while the apoptosis rate and expression of Bax and p21 were increased(P<0.05).Compared with the CUR-H group,the CUR-H+DMOG group exhibited increased angiogenic lumen formation,elevated Edu-positive rates,and higher expression of CyclinD1,Bcl-2,VEGFA,HIF-1α,VEGFR2,and p-AKT/AKT,whereas the apoptosis rate and expression of Bax and p21 were decreased(P<0.05).Conclusion Curcumin may inhibit proliferation and angiogenesis,and promote apoptosis in lung cancer cells,mechanisms that could be associated with the suppression of the HIF-1α/VEGFR/Akt pathway.关键词
