宫粘方通过提高子宫内膜容受性治疗宫腔粘连的机制研究
Study on the mechanism of Gongzhan Formula(宫粘方)in treating intrauterine adhesions by enhancing endometrial receptivity

Abstract
Objective To investigate the mechanism of Gongzhan Formula(宫粘方,GZF)in the treatment of intrauterine adhesions(IUA).Methods In vivo:Forty-five SD rats were randomly assigned to the sham surgery group(SHAM),amodel group(MOD),or the GZF group.Pathological changes in rat endometrial tissues were assessed by HE staining on postoperative days 1,3,5,10 and 28.The expression of integrin αvβ3 was evaluated by immunohistochemistry(IHC).In vitro:Endometrial stromal cells(ESCs)and macro-phages(M)were isolated and cultured from the endometrium of IUA model rats(n=3 per cell type).Cells cultured with serum contain-ing GZF were designated as the experimental groups(M+HYXQ and ESC+HYXQ).IF was used to detect the expression of cytokeratin and vimentin.CCK-8 assay was employed to assess ESC proliferation.Angiogenesis was evaluated using a tube formation assay.The mRNA expression of CD51 was measured by qRT-PCR.Results HE staining revealed significantly reduced gland number,increased inflammatory cell infiltration,and sparse and incomplete tissue structure in the MOD group endometrium compared to the SHAM group(P<0.05).In contrast,the GZF group showed alleviated inflammatory reaction,more complete and dense endometrial structure,and increased gland number(P<0.05).IHC demonstrated significantly higher expression of integrin αvβ3 in the GZF group compared to the MOD group(P<0.05).IF indicated significantly higher vimentin expression compared to cytokeratin in the endometrium of IUA model rats.In the ESC+HYXQ and M+HYXQ groups,stromal cell absorbance(indicating viability),proliferation rate,and the number of vascular lumens and nodes were significantly increased.Furthermore,CD51 mRNA expression was also significantly elevated in these groups(P<0.05).Conclusion GZF may promote gland formation,alleviate endometrial inflammation,and upregulate integrin αvβ3 expression,thereby enhancing endometrial stromal cell proliferation and angiogenesis.These effects potentially contribute to improved endometrial receptivity and the treatment of IUA.关键词
