Extraction of Fructus Ligustri Lucidi exosome-like nanovesicles and exploration of the anti-aging mechanism of Caenorhabditis elegans

Abstract
Objective To investigate the effect of Fructus Ligustri Lucidi exosome-like nanovesicles(FELNs)on the lifespan regulation of Caenorhabditis elegans(C.elegans)and preliminarily elucidate its molecular mechanism.Methods FELNs were extracted by ultra-centrifugation and characterized by transmission electron microscopy(TEM)and nanoparticle tracking analysis(NTA).Wild-type N2 C.elegans were used as the model organism,and different concentrations of FELNs were administered to intervention groups.The bio-logical effects were evaluated through lifespan assays,oxidative stress tests,and heat stress assays.Key genes and signaling pathways regulated by FELNs were analyzed using transcriptome sequencing.Results The successfully isolated FELNs exhibited a typical cup-shaped structure with an average particle size of 148.3 nm and were effectively taken up by the nematodes.Compared with the control group,treatment with 100 µg/mL FELNs significantly extended the maximum lifespan of the nematodes and markedly improved their sur-vival rates under oxidative and heat stress conditions.Transcriptomic analysis revealed 291 differentially expressed genes(DEGs)after FELNs intervention,primarily enriched in pathways such as neural signaling,innate immunity,and autophagy.Among these,genes including eat-4(glutamate transport regulation),col-10(collagen synthesis),and cpr-2(stress response)were significantly upregu-lated.Conclusion FELNs can significantly extend the lifespan of nematodes and enhance their stress resistance by multi-target regula-tion of neurotransmission,immune response,and autophagy pathways.This study provides new experimental evidence for the applica-tion of plant-derived nanovesicles in the field of anti-aging.关键词
