Abstract
Objectives:To investigate age-related characteristics of major testicular cell types and identify key molecular markers across developmental stages by analyzing published single-cell RNA sequencing(scRNA-seq)data of human testes using bioinformatics approaches.
Methods:The scRNA-seq datasets from human testicular tissues across different age groups were retrieved from the Gene Expression Omnibus database.Data analysis was performed using the Seurat toolkit.Cell types in each sample were annotated based on marker genes from multiple literature sources.
Results:Key molecular markers were identified for testicular cell types including Sertoli cells(SOX9,PRND,BEX1,BEX2,BEX5,CITED1),Leydig cells(DCN,IGF1,INHBA,LHX9,LUM),spermatogonial stem cells(POU5F1,NANOG,SOX2),spermatogonia(DND1,DMRT1,FGFR3,GFRA1,ID4,KIT,NANOS3,NR6A1,PIWIL4,SOHLH2,SSX3,UTF1),spermatocytes(CCNA1,DAZL,DMRTB1,MAGEA4,PIWIL1,PRDM9,PTTG1,SPATA16,SPO11,SYCP1,SYCP2,SYCP3,GPR85),and sperm cells(PRM1,PRM2,PRM3,RNF151,SPACA3,TNP1,TSSK6,TXNDC2).Further analysis found germ cell marker expression exhibited dynamic regulation in neonatal testes,during the prepubertal stage and after the onset of puberty.Additionally,Sertoli cells bifurcated into two distinct subgroups during early development,with one group highly expressing SOX9 and PRIND genes and gradually disappearing during development.
Conclusions:Based on literature-derived human testicular single-cell transcriptomic data,this study identified multiple specific marker genes of germ cells that enable accurate identification of cell types at different developmental stages.These findings provide a valuable foundation for understanding the developmental processes of the human reproductive system.关键词
