Inflammatory cytokines mediate the gut microbiota-autism spectrum disorder causal pathway:A Mendelian randomization study

Abstract
Objective To elucidate the causal association between gut microbiota and autism spectrum disorder(ASD)and quantified the mediating effects of inflammatory cytokines using multivariable mediation based on Mendelian randomization(MR).Methods Genome-wide Association Study summary data for gut microbiota,inflammatory cytokines,and ASD were utilized.MR was employed to investigate the causal relationship between gut microbiota and ASD,with the inverse variance weighting(IVW)method serving as the primary statistical approach.Additionally,mediation analysis was conducted to explore whether inflammatory cytokines mediated the pathway from gut microbiota to ASD.Results The forward MR analysis revealed a positive causal association between Prevotellamassilia and the risk of ASD(P<0.05),while reverse MR analysis showed no statistical significance(P>0.05).Sensitivity(MR-Egger regression)analysis confirmed the stability of the results.Mediation analysis indicated that the inflammatory cytokine natural killer cell receptor 2B4(NK2B4)partially mediated the causal relationship between Prevotellamassilia and ASD risk,with a mediation effect ratio of 7.06%.Conclusion The gut microbiota Prevotellamassilia exhibits a causal association with ASD,and inflammatory cytokine NK2B4 partially mediates this causal relationship.关键词
