Effect of Yanghe Pingchuan granules combined with bone marrow mesenchymal stem cell transplantation on polarization of M1/M2 macrophages in asthmatic rats

Abstract
Objective It is to study the effects of Yanghe Pingchuan granules combined with transplantation with bone marrow mesenchymal stem cells(BMSCs)on polarization of M1/M2 macrophages in asthmatic rats.Methods Forty-nine SD rats were randomly divided into 7 groups:normal group,model group,BMSCs group,BMSCs+low dose Yanghe Ping-chuan group,BMSCs+medium dose Yanghe Pingchuan group,BMSCs+high dose Yanghe Pingchuan group,and BMSCs+dexamethasone group,with 7 rats in each group.The rats of all groups except for the normal group were sensitized with ovalbumin and aluminum hydroxide to establish asthma models.During the asthma sensitized period,the BMSCs+low-,medium-,and high-dose Yanghe Pingchuan groups were given Yanghe Pingchuan granule suspensions 3.87 g/kg,7.74 g/kg,15.48 g/kg by gavage,respectively,the BMSCs+dexamethasone group was given dexamethasone suspension 0.025 mg/kg by gavage,all the other groups were given an equal volume of saline by gavage,and each BMSCs group was given BMSCs by intravenous injection from tail vein,all once daily,continuously treated for 6 weeks.The histopathological morphology of lung tissue was observed by HE staining,the levels of interleukin-4(IL-4),interleukin-13(IL-13)and Interferon-γ(IFN-γ)in bronchoalveolar lavage fluid were measured by ELISA,the protein expressions of Arginase 1(Arg-1)and in-ducible Nitric Oxide Synthase(iNOS)in lung tissue were detected by Western blot analysis,and the positive expressions of CD206 and CD86 in lung tissue were detected by immunohistochemical staining.Results Compared with the model group,the inflammatory cell infiltration in lung tissue was improved and the goblet cells and mucus were decreased in all medicated groups,the levels of IL-4 and IL-13 in bronchoalveolar lavage fluid,and the relative protein expression of Arg-1 in lung tis-sue,and mean optical density values of CD206-positive expression were all significantly decreased(all P<0.05),while the levels of IFN-γ in alveolar lavage fluid,relative protein expression of iNOS in lung tissue,and mean optical density val-ues of CD86-positive expression were significantly increased(all P<0.05).Compared with the BMSCs group,the levels of IL-4 and IL-13 in bronchoalveolar lavage fluid in the BMSCs+Yanghe Pingchuan groups and BMSCs+dexamethasone group,and the relative protein expression of Arg-1 in lung tissue,and mean optical density values of CD206-positive ex-pression were all significantly decreased(all P<0.05),while the levels of IFN-γ in alveolar lavage fluid,relative protein expression of iNOS in lung tissue,and mean optical density values of CD86-positive expression were significantly increased(all P<0.05).Conclusion Yanghe Pingchuan granules combined with BMSCs can inhibit M2 macrophage polarization and promote M1 macrophage polarization to restore the M1/M2 macrophage balance via down-regulating expressions of IL-4 and IL-13 and up-regulating expression of IFN-γ,thus to control airway inflammation in asthma.关键词
