Effect of Huchaoyin on apoptosis and autophagy of ovarian granulosa cells in rats with tripterygium wilfordii polyglycosides induced premature ovarian insufficiency

Abstract
Objective It is to explore the effect of Huchaoyin on apoptosis and autophagy of ovarian granulosa cells in rats with premature ovarian insufficiency(POI)induced by tripterygium wilfordii polyglycosides.Methods Fifty SD female rats with normal estrous cycle were divided into control group,model group,low-dose,medium-dose and high-dose groups of Huchaoyin,with 10 rats in each group.The rats of all groups except for the control group were gavaged with tripterygium wilfordii polyglycosides solution to establish models of POI.After modeling,the rats in the low,medium and high dose groups of Huchaoyin were gavaged with 5 g/kg,10 g/kg and 20 g/kg Huchaoyin respectively,while the control group and model group were gavaged with an equal amount of pure water,all once daily,continuously gavaged for 15 days.The es-trous cycles of rats in each group were monitored from the beginning of modeling to treatment period.After treatment,the serum levels of serum estradiol(E2),follicle-stimulating hormone(FSH),luteinizing hormone(LH)and anti-Mullerian hormone(AMH)in rats of each group were measured by ELISA,the pathological morphology of ovarian tissues was ob-served by HE staining,the apoptosis index of ovarian granulosa cells was analyzed by TUNEL,the positive expression of the autophagy marker microtubule-associated protein 1 light chain 3B(LC3B)in ovarian tissue were detected by immuno-histochemical staining,and the expressions of autophagy-related proteins LC3-Ⅰ,LC3-Ⅱ,and Beclin1 in ovarian tissue were detected by Western blot.Results Compared with the control group,the estrous cycle of rats in the model group was disordered and prolonged,the serum levels of E2 and AMH were significantly decreased,while the levels of FSH and LH were significantly increased(all P<0.05);the ovarian tissue structure was damaged,the arrangement of granulosa cells were disordered,the apoptosis index was significantly increased(P<0.05);the average optical density of LC3B and the ratio of LC3-Ⅱ/LC3-Ⅰin ovarian tissue were significantly increased,and the protein expression of Beclin1 was significant-ly upregulated(P<0.05).Compared with the model group,the estrous cycles of the rats in the low,medium and high dose groups of Huchaoyin groups were shortened,the serum levels of E2 and AMH were significantly increased,and the levels of FSH and LH were significantly decreased(all P<0.05),the ovarian tissue morphology was significantly im-proved,the apoptosis index of ovarian granulosa cells was significantly decreased(P<0.05);the average optical density of LC3B and the ratio of LC3-Ⅱ/LC3-Ⅰin ovarian tissue were significantly decreased,and the protein expression of Bec-lin1 was significantly down-regulated(P<0.05);the increase or decrease of these indicators were dose-dependent in the Huchaoyin group,and the differences were significant between the two groups(P<0.05).Conclusion Huchaoyin can reg-ulate the level of sex hormones,inhibit excessive autophagy of ovarian granulosa cells and reduce cell apoptosis,thereby improve ovarian function of rats with premature ovarian insufficiency.关键词
