摘要

Abstract

Objective It is to investigate the effect of Shumai capsule on renal damage and ferroptosis in diabetic rats.Methods Thirty SD rats were taken,in which 10 rats were randomly selected as control group and were fed normal diet(2 rats were excluded due to natural death),the remaining rats were fed with high-fat,high-sugar diet and given streptozotocin by intraperitoneal injection to establish models of diabetic kidney injury.16 successfully modeled rats were randomly divid-ed into model group and Shumai group,with 8 rats in each group.Starting from Day 1 after successful modeling,the Shu-mai group was given Shumai capsules 1.15 g/kg by gavage,the blank control group and model group were an equivalent volume of distilled water by gavage,all once daily,continuously treated for 28 days.The levels of blood glucose and renal function indicators,renal pathological changes,the contents of malondialdehyde(MDA),glutathione(GSH)and Fe2+in renal tissue,and the expressions of long-chain acyl-CoA synthase 4(ACSL4),solute carrier family 7 member 11(SLC7A11),glutathione peroxidase 4(GPX4),ferritin heavy chain 1(FTH1)and nucleolar assembly factor 1 ribonucle-oprotein(NAF1)in renal tissue were compared before and after treatment among the rats of every groups.Results On days 7,14,and 28 of gavage administration,the serum levels of FPG in rats of the model group were significantly higher than those in rats of the blank group at the same time points(all P<0.05),and the serum levels of FPG in rats of the Shumai group were significantly lower than those in rats of the model group(all P<0.05).Compared with the blank group,the serum levels of BUN and Cr in rats of the model group were significantly increased after the final gavage(all P<0.05);multiple areas of necrosis,inflammatory cell infiltration and hemorrhage were observed in the renal tissues;the contents of MDA and Fe2+,as well as the relative protein expression of ACSL4 in the renal tissue were significantly increased(all P<0.05),while the content of GSH and relative protein expressions of SLC7A11,GPX4,FTH1,and NAF1 in the renal tis-sue were significantly decreased(all P<0.05).Compared with the model group,the serum levels of BUN and Cr in rats of the Shumai group were significantly decreased after the final gavage(all P<0.05);the inflammatory cell infiltration in the renal tissue was decreased without obvious hemorrhage;the contents of MDA and Fe2+,as well as the relative protein expression of ACSL4 in the renal tissue were significantly decreased(all P<0.05),while the content of GSH and relative protein expressions of SLC7A11,GPX4,FTH1,and NAF1 in the renal tissue were significantly increased(all P<0.05).Conclusion Shumai capsule can obviously improve renal function and alleviate histopathological damage in rats with diabet-ic renal damage,alleviate oxidative stress and iron load in renal tissues,the mechanism of action may be related to regula-ting ferroptosis,reducing iron accumulation in renal tissue,and alleviating oxidative stress.
