健脾清热活血方对三硝基苯磺酸诱导溃疡性结肠炎大鼠HIF-1α、VEGF表达的影响
Effects of decoction for fortifying spleen,clearing heat and activating blood on expressions of HIF-1α and VEGF in rats with trinitrobenzenesulfonic acid-induced ulcerative colitis

Abstract
Objective It is to observe the effects of decoction for fortifying spleen, clearing heat and activating blood (DFSCHAB) on the colonic mucosal tissue structure and the expressions of hypoxia-inducible factor-1α (HIF-1α) and vascular endothelial growth factor (VEGF) in rats with trinitrobenzenesulfonic acid (TNBS)-induced ulcerative colitis, and to explore the potential mechanism of this formula in regulating vascular remodeling to prevent and treat ulcerative colitis.Methods Forty-eight clean grade male SD rats were taken and randomly divided into normal group, model group, DFSCHAB group and mesalazine group according to a weight ratio of 1: 1. The rats of all groups except for the normal group were given 5％ TNBS 100 mg/ kg +50％ ethanol by clyster to establish models of ulcerative colitis. After successful modeling, the DFSCHAB group was given DFSCHAB 6. 3 g/ kg (concentrated to 1 mL containing 1 g crude drug weight) by gavage, the mesalazine group was given mesalazine sustained-release granules 0. 4 g/ kg by gavage, both the model group and control group were an equal volume of physiological saline by gavage, all once daily, continuously treated for 2 weeks. the rats were divided into groups and intervened for 2 weeks. All rats were euthanized via intraperitoneal anesthesia at the end of the second week, a segment of intestine located 8 cm from the anus was resected, the histopathological morphology of the colon tissue was observed by HE staining, the ultrastructure of the colon tissue was observed by H-7650 transmission electron microscopy, the positive protein expressions of HIF-1α and VEGF in the colon tissue were detected by immunohistochemistry.Results HE staining showed that there were severe congestion and edema of the mucosa,partial epithelial shed-ding,multiple and large ulcers,and significant infiltration of inflammatory cells in the submucosal layer in the model group;there were mucosal epithelial shedding,congestion and swelling,small ulcer changes,and infiltration of inflamma-tory cells in the submucosal layer was alleviated in the DFSCHAB group and mesalazine group.Electron microscope obser-vation showed that there were vacuolization and autophagy changes in colonic epithelial cells of the model group,with obvi-ous focal necrosis,disordered arrangement of microvilli structures,and significant proliferation of mitochondria,Golgi ap-paratus,and ribosomes in the cells,localized expansion and swelling were also observed;there were decrease in volume and vacuolization of colonic epithelial cells in the DFSCHAB group and mesalazine group,with significant expansion and swelling of organelles,edge aggregation of nuclear chromatin,nuclear consolidation,nuclear membrane rupture,and visi-ble nuclear fragmentation,obvious formation of apoptotic bodies.HIF-1α and VEGF were high expressed in colonic muco-sal tissue in the model group,and their distribution areas were significantly higher than those in the normal group(all P<0.05);the expressions of HIF-1α and VEGF in colonic mucosal tissue were decreased in the DFSCHAB group and mesala-zine group,and their distribution areas were smaller than those in the model group(all P<0.05).Conclusion Decoction for fortifying spleen,clearing heat and activating blood can improve local tissue hypoxia and regulate autophagy balance in intestine by down-regulating the expressions of HIF-1α and VEGF,to promote mucosal healing in the treatment of ulcerative colitis.关键词
