花魔芋PAL基因家族的鉴定及其生物胁迫和非生物胁迫下的表达模式分析
Genome-wide identification and expression analysis of phenylalanine ammonia-lyase gene family under biotic and abiotic stresses in Amorphophallus konjac

Abstract
[Objective]Phenylalanine ammonia-lyase(PAL)is the key and rate-limiting enzyme in the phenylpropanoid pathway of plants,playing a critical role in growth,development and stress resistance.The study aimed to explore the characteristics and expression of AkPALs,which would provide genetic resources and references for in depth study of AkPAL gene function and molecular breeding of stress resistant konjaks.[Method]Based on the genomic information of the A.konjac,the AkPAL gene family members of A.konjac were accurately identified using local Blast analysis in TBtools and feature domain by referring to AkPAL family protein sequence of A.konjac.The physicochemical properties,gene structure,phylogenetic tree,conserved motifs,cis-acting elements and collinearity relationship of the AkPAL genes were ana-lyzed by bioinformatics methods.Moreover,their expression patterns in different tissues,at four growth stages of bulbs,and under the treatment with different biological stresses and abiotic stresses were compared and analyzed through transcriptome data.[Result]Nine PAL family genes were identified in the whole genome of A.konjac.PALs proteins had small differences in physicochemical properties and similar conserved mo-tifs and gene structures.Based on phylogenetic analysis,the AkPALs were divided into four clades,which were more closely related to PAL proteins in monocotyledonous pants(Oryza sativa and Zea mays).Multiple cis-acting elements related to multiple adversity stress response,phytohormone responsive and growth and development regulation and light response were present on the promoter of AkPALs.The AkPAL fam-ily genes were differentially expressed in different tissues,at different developmental stages of bulbs,and under the treatment of ABA(Ab-scisic acid),JA(Jasmonic acid),SA(Salicylic acid)and low-temperature and Pcc(Pectobacterium carotovorum subsp.Carotovorum).Ak-PAL03 and AkPAL04 genes were highly expressed in the root,bulb,petiole and leaf.Under different hormone treatments,AkPAL04 gene was up-regulated under the induction of ABA,JA and SA.Under low-temperature stress and pathogen Pcc stress,AkPAL04 gene was also signifi-cantly up-regulated.In addition,the AkPAL04 gene showed a relatively high expression level at all four stages of bulb development.[Conclu-sion]Nine PAL genes are obtained from A.konjac.Among them,AkPAL04 gene is highly expressed in all tissues of A.konjac and 4 develop-ment stages of bulbs,and is highly expressed under biotic and abiotic stress,suggesting that AkPAL04 may be involved in resistance to Pcc in-fection,ABA,SA,JA and low temperature in addition to regulating the growth and development of various tissues of A.konjac.AkPAL04 genes can be considered crucial candidates for stress response.关键词
