摘要

Abstract

[Objective]Sucrose transporters(SUTs)mediate sucrose transmembrane transport and play an important role in plant growth and development and stress.The purpose of the present study was to explore the function of StSUT2 regulating aluminum stress tolerance in pota-to.[Method]Transgenic lines of potato StSUT2 RNA interference expression and its wild type were used as materials,by analyzing the effects of StSUT2 interference expression on root tip cell wall ultrastructure,cell wall composition,pectin methylesterase(PME)activity and the ex-pression of related genes,the mechanism of StSUT2 regulating the resistance of potato to aluminum stress was elucidated.[Result]The rela-tive root elongation of StSUT2 interference line was significantly lower than that of the wild type under AlCl3 treatment,and more Al3+was accumulated in the root tips of StSUT2 interference line than that of the wild type,indicating that the tolerance of StSUT2 interference line to aluminum stress was significantly lower than that of the wild type,which meant that StSUT2 was involved in the positive regulation of alumi-num stress tolerance of potato.Further analysis showed that aluminum stress induced cell wall thickening and cell wall components cellulose,hemicellulose and lignin contents increased,but the cell wall thickness and cellulose,hemicellulose and lignin contents of StSUT2 interference lines were significantly higher than those of wild type before and after aluminum stress treatment.Under aluminum stress,the activity of pectin methylase in StSUT2 interfered lines was significantly higher than that of wild type,the expression of StPME gene was significantly higher than that of wild type,and the expression of StPEI gene was significantly lower than that of wild type.[Conclusion]StSUT2 interference ex-pression in potato decreases the resistance to aluminum stress.It is speculated that the intreference expression of StSUT2 changes the content of cell wall components,the thickness of cell wall and the activity of pectin methyl esterase in the root wall,which result in the accumulation of more Al3+in the root tips,thus affecting root elongation under aluminum stress.
