Abstract

Terahertz signals generated endogenously by cells are intrinsically weak and are easily masked by the high loss of background water.Accurate acquisition of terahertz information from living cells therefore demands highly sensitive detection techniques.Here a test platform is constructed based on electronic terahertz multi-band stitching technology to realize precise detection and analysis of tumor-cell solutions.Cells are resuspended in dimethyl sulfoxide(DMSO)as the background medium;this simultaneously preserves cell viability and eliminates water-related interference while amplifying the terahertz-response contrast between the cells and the surrounding medium.Using HeLa cells and human glioma cells(HS683)as model systems,solutions of both cell types at various concentrations are prepared firstly.The results show that the terahertz dielectric constant of the solutions increases monotonically with cell concentration,enabling discrimination of both cell types and concentrations via terahertz dielectric spectroscopy.Next,the physiological process of apoptosis is mimicked by exposing the cells to a water-bath treatment and terahertz complex dielectric spectra at different viability stages are obtained.These spectra reveal a progressive decrease in the terahertz dielectric constant as apoptosis proceeds.Finally,by correlating microscopic imaging with terahertz dielectric spectroscopy,the feasibility of using DMSO as a terahertz-compatible background for cell studies is verified and how the observed terahertz dielectric changes during apoptosis are linked to underlying microscopic morphological alterations is elucidated.
