Abstract

Objective To explore the potential bidirectional causal effect between immune cells and lung cancer,so as to provide scientific basis for clinical prevention and treatment of lung cancer.Methods The data of 731 immune cell characteristics in the GWAS directory and the lung cancer GWAS data of the International Lung Cancer Alliance were collected.The causal relationship between immune cell characteristics and lung cancer was evaluated by bidirectional two-sample Mendelian randomization analysis.The inverse variance weighting method was used,and heterogeneity test,sensitivity analysis and leave-one-out test were performed.Results After FDR adjustment,this study identified eight immunophenotypes associated with lung squamous cell carcinoma and lung adenocarcinoma(PFDR<0.05),including CD19 on IgD+CD38+B cells,CCR2 on granulocytes,percentage of naive CD8+T cells in CD8+T cells,absolute count of effector memory CD4-CD8-T cells,percentage of CD4+T cells in white blood cells,CD40 on monocytes,CCR2 on CD14+CD16+monocytes,and CD8 on naive CD8+T cells.For overall lung cancer,after adjusting for significance,two immunophenotypes were found to be associated with it:the absolute count of IgD+CD24-B cells and the percentage of TCRgdT cells in T cells.Reverse MR analysis showed that lung cancer had no statistically significant effect on immunophenotype.Conclusion This study confirms a potential causal relationship between immune cell traits and lung cancer.These findings provide new insights for clinical prevention and treatment of lung cancer.Changes in certain immune cell traits may influence the onset and progression of lung cancer,offering a theoretical foundation and research direction for developing immune-modulation-based intervention strategies for lung cancer.
