Abstract
AIM A high-performance liquid chromatography-tandem mass spectrometry(LC-MS/MS)method was established to detect the concentration of 3-indoxyl sulfate in the serum of children with acute lymphoblastic leukemia.METHODS Protein precipitation was employed to prepare the sample.A C18 column(4.6 mm x 150.0 mm,3.5 μm,Waters,USA)was utilized for the mobile phase.For gradient elution,a mixture of acetonitrile and a 10 mmol·L-1 ammonium acetate solution(volume ratio of 1∶9)was used.The flow rate was 0.8 mL·min-1,the column temperature was 25℃,and the sample volume was 5 μL.Using the electrospray ionization(ESI)in negative ion mode,the concentration of 3-indoxyl sulfate in serum samples of children with acute lymphoblastic leukemia was quantitatively detected.The quantitative ion pair for 3-indoxyl sulfate had a mass-to-charge ratio(m/z)of 212.0 →80.0.The declustering potential(DP)was set at-40 V,and the collision energy(CE)was-33 V.For hydrochlorothiazide,the quantitative ion pair had a m/z of 296.0 → 205.0,with a DP of-48 V and a CE of-33 V.Additionally,the ion source temperature was maintained at 600℃.RESULTS The linear relationship of 3-indoxyl sulfate in serum samples from children with acute lymphoblastic leukemia was good in the range of 0.250 0 to 10.000 0 mg·L-1.The lower limit of quantification was established at 0.250 0 mg·L-1.Accuracy(the corresponding RE)within and between batches was found to be 1.320％-2.140％and 1.460％-3.470％,respectively,while the precision(the corresponding RSD)within and between batches was found to be 3.160％-5.500％and 2.990％-5.170％,respectively.The average extraction recovery rate ranged from 86.40％to 95.60％,while the matrix effect varied between 91.73％and 94.60％.CONCLUSION The method established in this experiment is simple,sensitive,and accurate,which is suitable for determining 3-indoxyl sulfate in the serum of children with acute lymphoblastic leukemia.关键词
