Abstract
To develop a feline calicivirus virus-like particle(FCV-VLP)vaccine with favorable safety and immunogenicity,this study constructed the pSMA-FCV-VP1 expression vector encoding the FCV capsid protein VP1.The recombinant His-SUMO-VP1 fusion protein was obtained by induction in E.coli BL21(DE3),followed by SUMO protease cleavage in the assembly buffer to remove the His-SUMO tag and fa-cilitate FCV-VLP assembly.The obtained FCV-VLPs were identified by SDS-PAGE,Western-blot,nanoparti-cle size analyzer and transmission electron microscope.The results showed that the VP1 band size was consistent with the expected target band size(66 kDa),and the assembled VLPs had a uniform particle size(30-40 nm),which was consistent with the size of feline calicivirus particles.After immunizing mice with FCV-VLPs,the level of feline calicivirus specific antibody in seram was 1∶720,and the virus-like particles could effectively promote the proliferation of spleen lymphocytes in mice.The FCV-VLPs pre-pared in this study can induce strong immune response,which lays a good foundation for vaccine devel-opment against feline calicivirus disease.关键词
