融合禽腺病毒4型和8a型Hexon蛋白亚单位疫苗免疫效果的评价
Evaluation of fusion fowl adenovirus type 4 and 8a Hexon protein subunit vaccine immunization effect

Abstract
In order to prepare a subunit multivalent vaccine against fowl adenovirus type 4(FAdV-4)and 8a(FAdV-8a),FAdV-4 and FAdV-8a Hexon protein fragments and their bivalent fusion proteins were ex-pressed to prepare the vaccine and evaluate the immunization effect.The expression and immunogenicity of the recombinant proteins were identified by SDS-PAGE and Western-blot,respectively.The recombi-nant proteins were used to prepare the vaccines for animal immunization.The immunized animals were challenged with FAdV-4 and FAdV-8a,respectively.The levels of serum specific antibody secretion and the expression levels of IL-2,IFN-γ,IL-4 and IL-6 were detected by ELISA;and the experimental animals were dissected and examined for disease.And the experimental animals were observed by dissection le-sions and histopathology.The results showed that the constructed recombinant proteins were expressed in the form of inclusion bodies and could react specifically with fowl adenovirus-positive serum;the specific antibodies induced by the subunit vaccines were significantly higher than those of the con-trol group(P<0.01),and the levels of cytokines IL-2,IFN-γ,IL-4,and IL-6 induced by the subunit vac-cine were significantly higher than those of the control group(P<0.01).After autopsy,some pericar-dial effusions and liver tissues of chickens in the challeged group were found to have different degrees of hemorrhages and yellow staining,while the livers of chickens in the immunized group showed no significant changes compared with those of the control group;histopathological observations showed that basophilic inclusion bodies and intercellular inflammatory cellular infiltration were observed in the hepatocytes of the challeged group,whereas no obvious lesions were observed in the immunized group.The present study demonstrated that the constructed fowl adenovirus type 4 and 8a Hexon bivalent fusion protein vaccines were able to induce a high level immune response to increase the protection rate of chicks and improve the liver tissue damage caused by the virus infections,which can provide a basis for the research of prevention and control of fowl adenoviruses.关键词
