Bioinformatics and Cadmium Response Characteristics Analysis of Sugar Beet BvMT2 Gene

Abstract
[Objective]Metallothioneins(MT),widely distributed metal-binding proteins in organisms,play crucial roles in plants'resistance to heavy metal stress.This study aimed to investigate the response characteristics of sugar beet metallothionein 2(BvMT2)under cadmium(Cd)stress.[Methods]The gene was cloned using RT-PCR technology,and bioinformatics methods were employed to predict cis-acting elements,upstream transcription factors,and physicochemical properties of the gene.Its transcriptional expression under varying Cd stress concentrations was analyzed.[Results]The coding region of the gene is 225 bp,with a relative molecular weight of 1 909.91 Da.It has a theoretical isoelectric point of 4.55 and is characterized by high contents of cysteine(Cys)and glycine(Gly),classifying it as an unstable,non-secretory,hydrophilic protein likely localized in the nucleus or chloroplasts.BvMT2 contains seven relevant cis-acting elements including CGTCA-motif,auxin related,light-response,partial light-responsive elements with conserved sequences,MYB-binding,anaerobic induction,and regulation.Transcription factors BVRB_7g177640 and BVRB_3g048050 are implicated in the transcriptional regulation of BvMT2.Transcriptome data indicated significant upregulation of BvMT2 under Cd stress at concentrations of 1,3 and 5 mmol/L.[Conclusion]In conclusion,BvMT2 plays a crucial role in sugar beet's response to Cd stress.This study establishes a foundation for understanding the function of BvMT2 in responding to heavy metal stress and provides insights into the mechanisms of sugar beet response to heavy metal pollution.关键词
