Abstract

Objective This study aims to explore the expression of IDO1 in human tumor tissues and its role in the tumor microenvironment.Methods The role of IDO1 in the pathogenesis of different tumors was explored in terms of gene expression,survival prognosis and immune infiltration using the cancer genome atlas(TCGA)and a combination of online sites such as GEPIA2,TIMER2.0 and LinkedOmics.GEPIA2 databases were used to obtain IDO1-related genes for gene ontology(GO)and kyoto encyclopedia of genes and genomes(KEGG)analysis.In addition,GO and KEGG enrichment analysis were performed on the protective mechanism of IDO1 in skin cutaneous melanoma(SKCM).Results Compared with normal tissues,IDO1 expression was up-regulated in 13 tumor tissues.Cox regression analysis showed that high expression of IDO1 was associated with poor prognosis in the majority of tumor patients.However,in skin cutaneous melanoma(SKCM)and head and neck squamous cell carcinoma(HNSC),high expression of IDO1 showed a good prognosis in terms of overall survival(OS)and disease-specific survival(DSS).Immune infiltration analysis revealed that IDO1 showed a strong positive correlation with M1 macrophages,but a negative correlation with a variety of immune cell infiltration.KEGG pathway enrichment analysis showed that IDO1 were mainly involved in the regulation of immune responses.In SKCM,KEGG pathway enrichment analysis focused on"antigen processing and presentation"and"Th1 and Th2 cell differentiation".Conclusions IDO1 expression is associated with clinical prognosis and immune cell infiltration in a variety of tumor tissues.IDO1 is the occurrence and development of tumors by influencing the body's immune response.
