Abstract

Objective To investigate the lipid-lowering effect and safety of Shanhe Jiangzhi Mixture in the treatment of hyperlipidemia.Methods High-performance liquid chromatography(HPLC)method was used for the content determination of aurantio-obtusin and chrysophanol in the preparation,the chromatographic column was Shimadzu-GL Inertsil® ODS-3 RP C18 column(250 mm×4.6 mm,5 µm),the mobile phase was 0.1%phosphoric acid aqueous solution-acetonitrile(gradient elution),the flow rate was 1.0 mL/min,the detection wavelength was 284 nm,the column temperature was 40℃,and the injection volume was 10 µL.Forty specific pathogen-free(SPF)KM mice were randomly divided into the control group(purified water)and the Shanhe Jiangzhi Mixture group(90 g/kg,calculated by raw drugs),and 0.4 mL of the corresponding drug was administered orally for every 10 g body weight.The body weight and heart,liver,spleen,lung,and kidney coefficients were detected.Sixty SPF SD rats were divided into the blank control group(group A,10 rats)and the model group(50 rats).The rats in the model group were orally administered 1.5 mL of sodium deoxycholate-lard-cholesterol propylene glycol-Tween 80(1∶6∶3∶1.2∶2.4,V/V/V/V)per 100 g body weight,while the rats in group A were orally administered an equal volume of ultrapure water once a day for 14 d to replicate the hyperlipidemia rat model.Forty-six successfully modeled rats in the model group were randomly divided into the model control group(group B,10 rats),the fenofibrate positive control group(group C,100 mg/kg),the low-,medium-,and high-dose groups of Shanhe Jiangzhi Mixture(groups D1,D2,D3,calculated as 5.25,10.50,21.00 g/kg for the original and raw drugs,respectively),with nine rats in each group.The rats in groups C,D1,D2,and D3 were given corresponding drugs by gavage daily,while rats in groups A and B were given an equal volume of 0.5%carboxymethyl cellulose sodium(CMC-Na)solution by gavage once a day for 14 d.The body weight of rats before and after the administration was measured,and serum triglycerides(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-C),and low-density lipoprotein cholesterol(LDL-C)levels were measured by colorimetric method.Very low-density lipoprotein cholesterol(VLDL-C)levels were measured by the fully automated biochemical analyzer,and TC/HDL-C ratio was calculated.Results The linear ranges of quercetin and emodin were 4.10-262.46 µg/mL and 4.01-256.45 µg/mL(R2=0.999 4,0.999 3,n=7),respectively.The RSDs of precision,stability,and repeatability test results were all lower than 2.0%.The average recoveries of quercetin and emodin were 94.09%and 90.60%,with RSDs of 7.32%and 7.60%(n=9),respectively.The contents of quercetin and emodin in the preparation were 87.66 and 151.16 µg/mL,respectively.The acute toxicity test showed that the heart coefficient in the Shanhe Jiangzhi Mixture group was significantly lower than that in the control group(P<0.01),while there were no significant differences in other indexes(P>0.05).The lipid-lowering effect study showed that the levels of serum TG,TC,HDL-C,and LDL-C in group C were significantly lower than those in group B(P<0.05),the serum LDL-C level and TC/HDL-C ratio in group D1 and D2 were significantly lower than those in group B(P<0.05),and the serum LDL-C level in group D3 was significantly lower than those in group B(P<0.05),while the serum LDL-C level in group D3 was significantly higher than that in groups D2 and D1.The levels of serum TC and HDL-C in groups D1,D2,and D3 were significantly higher than those in group C(P<0.05).Conclusion This method is simple,stable,and has good reproducibility.This mixture has a certain lipid-lowering effect and no significant acute toxicity,which can be used to treat hyperlipidemia.
