蒙药诃子对乌头碱诱导心脏毒性模型斑马鱼的保护作用
Protective Effect of Mongolian Medicinal Herb Terminaliae Belliricae Fructus Against Aconitine-Induced Cardiotoxicity in the Zebrafish Model

Abstract
Objective To investigate the protective effect of Terminaliae Belliricae Fructus(TBF)against aconitine-induced cardiotoxicity in the zebrafish model.Methods TBF freeze-dried powder was prepared,and the optimal dosages of TBF and aconitine for modeling were determined through the tolerance tests.Zebrafish fertilized for 6 h(6 hpf)were divided into the blank control group(group G,Holt-Buffer culture drugs),the model group(group H,10 µmol/L aconitine),and low-,medium-,and high-dosage groups of TBF freeze-dried powder(groups I,J,K,10 µmol/L aconitine+7.5,15.0,30.0 µmol/L TBF freeze-dried powder,calculated based on the content of raw drugs).After 48 h of incubation,the heart rate and cardiovascular morphology changes of zebrafishes were observed.The zebrafishes were collected and freezed,and the enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of myocardial injury-related factors[creatine kinase-MB isoenzyme(CK-MB)and troponin(Tn)],and inflammatory factors[tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β)],and oxidative stress-related factors[malondialdehyde(MDA)and superoxide dismutase(SOD)].Results The heart injury of zebrafishes in group H was significant,but the heart injury of zebrafishes in groups I,J,and K improved to varying degrees,and pericardial edema significantly reduced.Compared with those in group H,the heart rate of zebrafishes in groups I,J,and K significantly slowed down(P<0.01);the levels of CK-MB,Tn,IL-1β,TNF-α,and MDA in groups I,J and K of zebrafishes significantly decreased(P<0.05);the SOD level of zebrafishes in group K significantly increased(P<0.01).Conclusion TBF has a certain zebrafishes protective effect on the aconitine-induced cardiotoxicity in the zebrafish model,and can significantly improve its cardiovascular morphology,heart rate,and levels of heart injury-related factors.关键词
