Network Pharmacology and Molecular Docking Study on the Mechanism of Jinhong Decoction in Treating Acute Lung Injury Induced by Sepsis

Abstract

Objective:To explore the mechanism of Jinhong Decoction in treating acute lung injury(ALI)in-duced by sepsis using network pharmacology and molecular docking techniques.Methods:Active compounds and targets of Jinhong Decoction were identified through databases including TCMSP,SwissADME,UniProt,GeneCards,OMIM,as well as literature mining.An"herb-compound-target"network was constructed using Cytoscape 3.9.1.Venn diagrams were used to identify key overlapping targets between Jinhong Decoction and sepsis-induced ALI.Protein-protein interaction(PPI)networks were analyzed via the STRING 11.5 database.Gene Ontology(GO)en-richment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses were performed using the DA-VID database.Molecular docking between active components of Jinhong Decoction and core targets was validated and visualized using the TCMSPAS system and Pymol software.Results:A total of 144 potential targets of Jin-hong Decoction and 1 416 disease-related targets for sepsis-induced ALI were identified.Twenty-three common targets were obtained through intersection analysis,with core targets including TNF,IL-1β,TP53,CXCL8,MAPK3,and Alb.Enrichment analysis revealed 364 GO terms and 106 KEGG pathways.Key signaling pathways included the MAPK,Toll-like receptor,IL-17,and TNF signaling pathways.Molecular docking results indicated strong binding affinities between the main target TNF and active compounds such as chrysin,luteolin,and querce-tin.Conclusion:Jinhong Decoction exerts therapeutic effects on sepsis-induced ALI through multiple compo-nents,targets,and pathways,providing a theoretical basis and reference for clinical prevention and treatment.
