基于cGAS/STING信号通路探究加味黄芪桂枝五物汤对RA炎症反应与滑膜增生的影响
Investigating the Effects of Modified Huangqi Guizhi Wuwu Decoction on Inflammatory Response and Synovial Hyperplasia in Rheumatoid Arthritis via the cGAS/STING Signaling Pathway

Abstract
Objective:To investigate the effects of Modified Huangqi Guizhi Wuwu Decoction on inflammatory response and synovial hyperplasia in rheumatoid arthritis(RA)based on the cyclic GMP-AMP synthase(cGAS)/stimulator of interferon genes(STING)signaling pathway.Methods:An in vitro RA model was established by stimulating MH7A human fibroblast-like synoviocytes(FLS)with lipopolysaccharide(LPS).Cells were divided in-to the control group,model group,and low-,medium-,and high-dose Modified Huangqi Guizhi Wuwu Decoction groups.Cell viability was assessed using the CCK-8 assay.Cell apoptosis was detected by Annexin V-FITC/PI double staining.Enzyme-linked immunosorbent assay(ELISA)was used to measure the levels of tumor necrosis factor-α(TNF-α),interferon-β(IFN-β),and interleukin-1β(IL-1β)in cell supernatants.Western blotting was performed to detect protein expression of cGAS and STING,and real-time PCR(RT-PCR)was used to determine their mRNA expression levels.Results:CCK-8 results showed that the optimal concentrations of Modified Huangqi Guizhi Wuwu Decoction for inhibiting cell proliferation were 0.05,0.1,and 0.5 mg/mL(low-,medium-,and high-doses),with a 24-h treatment duration selected for subsequent experiments.Flow cytometry analysis of apoptosis revealed that,compared with the control group,the apoptosis rate significantly decreased in the model group;compared with the model group,apoptosis rates significantly increased in the low-,medium-,and high-dose Modified Huangqi Guizhi Wuwu Decoction groups.ELISA results showed that,compared with the control group,TNF-α,IFN-β,and IL-1β levels were upregulated in the model group;compared with the model group,these cytokines were significantly downregulated in the Modified Huangqi Guizhi Wuwu Decoction groups(P<0.01).Western blotting results indicated that cGAS and STING protein expression were upregulated in the model group compared with the control group,and downregulated in the Modified Huangqi Guizhi Wuwu Decoction groups compared with the model group(P<0.01).RT-PCR results showed that cGAS and STING mRNA expression fol-lowed the same trend:upregulated in the model group and downregulated in the Modified Huangqi Guizhi Wuwu Decoction groups(P<0.01).Conclusion:Modified Huangqi Guizhi Wuwu Decoction can modulate inflammatory re-sponses and abnormal proliferation in MH7A human fibroblast-like synoviocytes,and its mechanism may be associ-ated with the cGAS/STING signaling pathway.关键词
