摘要

Abstract

Objective:To observe the effects of stellate ganglion embedding therapy on airway remodeling and lung tissue endothelin-1(ET-1)levels in a rat model of asthma,and to observe its therapeutic efficacy and mecha-nism in modulating airway remodeling.Methods:40 male Sprague-Dawley(SD)rats were randomly divided into five groups:blank control group,model group,stellate embedding(star-embedding)group,sham-embedding group,and hormone(positive control)group,with 8 rats in each group.Except for the blank group,asthma models were established in the other groups using ovalbumin(OVA)sensitization and challenge.The star-embedding group received stellate ganglion embedding treatment once weekly for 14 days;the sham-embedding group under-went identical procedures but without leaving the suture in place.The hormone group received oral gavage of glu-cocorticoids for 14 days.Clinical manifestations and respiratory rates were observed and scored.Hematoxylin-eosin(H&E)staining and periodic acid-Schiff(PAS)staining were used to evaluate morphological changes in lung tis-sues,including epithelial shedding and goblet cell hyperplasia.Enzyme-linked immunosorbent assay(ELISA)was performed to measure levels of ET-1 and matrix metalloproteinase-9(MMP-9)in bronchoalveolar lavage fluid(BALF).Western blotting was used to detect ET-1 protein expression in lung tissues.Results:After intervention,respiratory rates in the star-embedding and hormone groups were significantly lower than in the model group(P<0.01),while no significant reduction was observed in the sham-embedding group(P>0.05).Lung tissues in the blank group showed normal airway structure with orderly arranged bronchial ciliated epithelium.In contrast,the model group exhibited thickened airway walls,narrowed lumens,increased mucus secretion,widened alveolar sep-ta,and inflammatory cell infiltration around bronchioles and in lung interstitium.Epithelial shedding(P<0.01)and goblet cell hyperplasia(P<0.01)were significantly increased.Both the star-embedding and hormone groups showed improved airway architecture,with obvisous alleviation of epithelial shedding and goblet cell hyperplasia compared to the model group(P<0.05).Levels of ET-1 and MMP-9 in BALF were significantly higher in the model group than in the blank group(P<0.01).Compared with the model group,both ET-1 and MMP-9 levels de-creased in the star-embedding group(P<0.05),though not as obvious as in the hormone group.ET-1 protein ex-pression in lung tissue was significantly upregulated in the model group(P<0.01).The hormone and star-embed-ding groups showed significantly reduced ET-1 expression compared to the model group(P<0.01),while no signif-icant change was observed in the sham-embedding group.Conclusion:Stellate ganglion embedding therapy ex-erts therapeutic effects in asthmatic rats by effectively reducing respiratory rate,improving respiratory function,and delaying airway remodeling.Its mechanism may be associated with downregulation of ET-1 protein expression in lung tissues.
