刺血疗法调控急性痛风性关节炎大鼠炎症反应的机制研究
Mechanism of Blood-letting Puncturing in Regulating Inflammatory Response in Rats with Acute Gouty Arthritis

Abstract
Objective:To investigate the mechanism of blood-letting puncturing on the inflammatory response of acute gouty arthritis rats.Methods:40 male Sprague-Dawley(SD)rats were randomly divided into four groups:blank control group,model group,blood-letting puncturing group,and colchicine group,with 10 rats in each group.Joint swelling degree and gait scores were assessed.Enzyme-linked immunosorbent assay(ELISA)was used to measure serum levels of interleukin-18(IL-18),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),myeloperoxidase(MPO),and blood uric acid(BUA).Histopathological changes in synovial tissue were examined using hematoxylin-eosin(HE)staining.Immunofluorescence staining was performed to detect neutrophil elastase(NE)and MPO protein expression in synovial tissue.Western blotting was used to assess protein expres-sion of NE,MPO,caspase-1,NOD-like receptor family pyrin domain-containing 3(NLRP3),and apoptosis-associ-ated speck-like protein containing a CARD(ASC)in synovial tissue.Results:At 6,12,and 24 hours after model-ing,the model group showed significantly higher ankle joint swelling rate,gait scores,serum levels of IL-18,IL-1β,TNF-α,MPO,and BUA,as well as increased protein expression of MPO,NE,caspase-1,NLRP3,and ASC in synovial tissue compared with the blank control group(P<0.05).Compared with the model group,both the blood-letting puncturing and colchicine groups exhibited significantly reduced ankle joint swelling,improved gait scores,lower serum levels of IL-18,IL-1β,TNF-α,MPO,and BUA,and decreased expression of MPO,NE,caspase-1,NLRP3,and ASC proteins in synovial tissue(P<0.05).Conclusion:Blood-letting puncturing may alleviate in-flammatory responses during acute gouty arthritis attacks by inhibiting NLRP3 inflammasome activation and there-by suppressing the formation and release of neutrophil extracellular traps(NETs).关键词
