参附注射液联合标准操作程序对脓毒症休克大鼠心肌抑制作用的研究
Study on the Myocardial Inhibitory Effects of Shenfu Injection Combined with Standard Operating Procedure in Rats with Septic Shock

Abstract
Objective:To investigate the mechanism of Shenfu injection combined with standard operating proce-dure(SOP)in alleviating myocardial inhibition in rats with septic shock.Methods:60 Sprague-Dawley(SD)rats were randomly divided into five groups,12 in each group.A myocardial inhibition model was established by intra-peritoneal injection of lipopolysaccharide(LPS)at a dose of 10 mg/kg.Model success was confirmed by measuring serum levels of cardiac troponin l(cTnl)and N-terminal pro-B-type natriuretic peptide(NT-proBNP),as well as by echocardiographic assessment.The successfully modeled rats were then randomly assigned to the sham group,model group,Shenfu group,SOP group,or Shenfu+SOP group,12 in each group.Hematoxylin-eosin(HE)stain-ing was used to evaluate myocardial pathological changes.Enzyme-linked immunosorbent assay(ELISA)was per-formed to measure serum levels of NT-proBNP,interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).West-ern blotting was used to detect protein expression of phosphorylated p38 mitogen-activated protein kinase(p-p38MAPK)and total p38MAPK.Results:After 3 and 6 h of treatment,the heart rate(HR),respiratory rate(R),and mean arterial pressure(MAP)in the Shenfu+SOP group were significantly improved compared with the mod-el group and the Shenfu group(P<0.05).After 6 h of treatment,the IL-6 levels in the Shenfu+SOP group were significantly lower than those in the SOP group(P<0.05).Severe inflammatory reactions,including myocardial de-generation and necrosis,were observed in all septic shock groups.Electron microscopy revealed nuclear pyknosis,reduced mitochondria,and interstitial fibrosis in cardiomyocytes.The expression levels of p-p38MAPK and p38MAPK in the Shenfu+SOP group showed statistically significant differences compared with the SOP group at the same time points(P<0.05).Conclusion:Shenfu injection combined with SOP can effectively alleviate myocar-dial inhibition in rats with septic shock.关键词
