基于生物信息学/网络拓扑策略探讨针灸治疗脊髓损伤的作用机制
Mechanistic Insights into Acupuncture and Moxibustion Therapy for SCI Using Bioinformatics and Network Topology Approaches

Abstract
Objective:To explore the molecular targets and mechanisms of acupuncture and moxibustion therapy in treatment of spinal cord injury(SCI)by using bioinformatics and network topology approaches.Methods:Major databases,including CNKI and PubMed,were systematically searched to identify the targets of acupoints.SCI-related targets were retrieved from databases such as GeneCards,and shared targets between acupoints and SCI were identified using the Venny website.Protein-protein interaction(PPI)networks and'acupoint-shared target'interaction networks were constructed using the String database and Cytoscape software,respectively.GO and KEGG enrichment analyses of the shared targets were performed using the Metascape database.Results:A total of 111 shared targets between acupoints and SCI were identified,with key targets including IL6,TNF,IL1B,STAT3,TGFB1,EGFR,BCL2,FGF2,HIF1A,and TLR4.GO and KEGG pathway enrichment analyses indicated that these targets mainly influence cell differentiation,programmed cell death,and miRNA regulatory processes,and were involved in signaling pathways such as NLR,MAPK,PI3K-AKT,and necroptosis.Conclusion:Acupuncture and moxibustion therapy may act on core targets such as IL6,TNF,IL1B,and STAT3,modulating the'neuroinflammation-programmed cell death'pathological network through multiple signaling pathways,including necroptosis,NLR,MAPK,and PI3K-AKT,thereby improving neurological and motor function after SCI.关键词
