摘要

Abstract

Objective:To observe the effects of electro-acupuncture(EA)at Neiguan(PC6)acupoint with three different current intensities(0.5 mA,1 mA,and 2 mA)on myocardial ischemia-reperfusion injury(MIRI)in rats,and to explore the underlying mechanism of ferroptosis regulation via the Nrf2-GPX4 signaling pathway.Methods:A total of 60 SPF-grade male SD rats were randomly divided into the Sham group,the ischemia-reperfusion(IR)group,the 0.5 mA group,the 1 mA group,and the 2 mA group,with 12 rats in each group.The MIRI model was established by ligating the left anterior descending coronary artery for 30 minutes followed by 24 hours of reperfusion.In the Sham group,the needle was passed through without ligation.Rats in the three EA groups received intervention at PC6 with different current intensities immediately at the onset of reperfusion.After 24 hours of reperfusion,the myocardial infarct size was assessed using TTC staining.Serum levels of myocardial enzymes(CK-MB,CK,and LDH)and inflammatory cytokines(TNF-α,IL-10,IL-6,and IL-1β)were measured by ELISA.Myocardial tissue oxidative stress markers(ROS,MDA,SOD,and GSH)were also evaluated.Ferrous ion(Fe2+)levels were measured using Fe2+assay kit.Protein expression levels of Nrf2 and ACSL4 in myocardial tissues were determined by Western blot.Results:Compared with the Sham group,rats in the IR group exhibited a significant increase in myocardial infarct size,serum levels of CK-MB,CK,and LDH,myocardial contents of ROS,MDA,Fe2+,and ACSL4 protein expression(P<0.05);the levels of inflammatory cytokines TNF-α,IL-6,and IL-1β were significantly increased(P<0.01);the levels of antioxidant markers SOD and GSH,and the protein expressions of Nrf2,GPX4,and FTH1 were significantly decreased(P<0.05,P<0.01);the level of IL-10 was markedly reduced(P<0.01).Compared with the IR model group,all three EA groups showed significant reductions in myocardial infarct size,serum levels of CK-MB,CK,and LDH,myocardial contents of ROS,MDA,Fe2+,and ACSL4 protein expression(P<0.05);the levels of inflammatory cytokines TNF-α,IL-6,and IL-1 βwere markedly decreased(P<0.01);the levels of SOD and GSH,and protein expressions of Nrf2 and GPX4 were significantly increased(P<0.05,P<0.01);the level of IL-10 was also significantly elevated(P<0.01).Conclusion:Electro-needling PC6 with current intensities of 0.5 mA,1 mA,and 2 mA all showed protective effects against MIRI.The underlying mechanism may involve activation of the Nrf2-GPX4 signaling axis,thereby inhibiting ferroptosis.Among the three,the 2 mA current group demonstrated the most effective improvement in MIRI.
