摘要

Abstract

Objective:To investigate the analgesic effect of electro-needling Baliao acupoints on a chronic anal rectal pain(CAP)rat model,and to explore its impact on the spinal p38 MAPK/TRPV1 signaling pathway,in order to elucidate the underlying mechanisms of electro-acupuncture(EA)in treatment of CAP.Methods:The CAP model was established via intracolonic instillation of capsaicin in SD rats.After successful modeling,the rats were randomly divided into the model group and the EA group,with 6 rats in each group;an additional 6 SD rats served as the blank control.The EA group was treated with electro-needling Baliao acupoints for 30 minutes daily over 7 consecutive days.Visceral pain-related behavioural responses and mechanical pain thresholds were assessed.HE staining was used to observe changes in the submucosal tissue of the rectum.Immunofluorescence was used to detect microglial activation and the expressions of P2X4R and TRPV1.Western blot analysis was conducted to measure the expressions of pathway-related proteins including P2X4R,phosphorylated p38 MAPK(p-p38),and TRPV1.Results:Compared with the blank group,the model group showed significantly stronger visceral pain-related behavioural responses and mechanical stimulation responses,with evident activation of microglial cells;the differences were statistically significant(P<0.01).In the EA group,these indicators were significantly improved compared to those in the model group,with statistically significant differences(P<0.01).In comparison to the blank group,the expressions of TRPV1 and P2X4R in the spinal cords were significantly increased,and the p-p38 was notably activated in the model group,with statistical significance(P<0.05).EA treatment suppressed the expression of TRPV1,P2X4R,and p-p38,with statistical significance(P<0.01).Conclusion:Electro-needling Baliao points may relieve CAP pain by regulating spinal p38MAPK/TRPV1 signaling pathway.
