茅莓水提物抗大鼠肝纤维化的作用及机制研究
Study on the effect and mechanism of aqueous extract of Rubus parvifolius L.on anti-hepatic fibrosis

Abstract
Objective To investigate the effect and mechanism of aqueous extract of Rubus parvifolius L.(ERPL)on liver fibrosis.Methods Twenty-four male SD rats were randomly divided into four groups(6 per group):control group,model group,ERPL low-dose group,and ERPL high-dose group.Except for the control group,the other three groups were subcutaneously injected with carbon tetrachloride(CCl4)twice a week for 8 weeks to establish a liver fibrosis model.Starting from the 5th week of modeling,the ERPL low-dose and high-dose groups were administered ERPL via gavage,while the control and model groups received 0.9%sodium saline by gavage,2 mL per rat,once a day,for 8 weeks.Blood was collected from the orbit and liver tissues of rats were collected.Liver tissue pathological changes in each group were observed using hematoxylin-eosin staining and Masson staining.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),laminin(LN),procollagen Ⅲ N-terminal peptide(PⅢNP),and collagen type Ⅳ(Col-Ⅳ)were measured by enzyme-linked immunosorbent assay.The relative protein expression levels of p53,solute carrier family 7 member 11(SLC7A11),and glutathione peroxidase 4(GPX4)in liver tissues were detected by Western blot.Results Compared with the model group,the ERPL low-and high-dose groups showed significantly reduced inflammatory infiltration and liver fibrosis in the liver tissue portal area of rats,as well as markedly decreased levels of ALT,AST,ALP,and fibrosis-related proteins LN,PⅢNP,and Col-Ⅳ(all P<0.05).In addition,the relative expression of p53 protein in liver tissue was significantly increased,while the protein levels of GPX4 and SLC7A11 were significantly decreased in the ERPL-treated groups compared with the model group(all P<0.05).Conclusion ERPL induced ferroptosis response by regulating the p53/SLC7A11/GPX4 pathway,thereby ameliorating liver fibrosis.关键词
