基于FAERS数据库的抗血小板类药物支架内血栓形成不良事件风险分析
Risk analysis of adverse events related to in-stent thrombosis of antiplatelet drugs based on the FAERS database

Abstract
Objective To mine and analyze adverse event reports of in-stent thrombosis associated with antiplatelet drugs based on the United States Food and Drug Administration Adverse Event Reporting System(FAERS)database so as to provide a reference for safe and rational clinical medication.Methods Adverse event reports of in-stent thrombosis associated with antiplatelet drugs recorded in FAERS from October 1,2003,to December 31,2024,were extracted.The OpenVigil 2.1 data platform analysis tool was used to mine and analyze risk signals employing the reporting odds ratio(ROR)method and the proportional reporting ratio(PRR)method.Results A total of 939 adverse event reports of in-stent thrombosis were retrieved from the FAERS database with antiplatelet drugs as the primary suspect medication.Among cases the proportion of males(58.47%)was higher than that of females(17.78%).The majority of patients experiencing adverse events were aged between 40-64 years(31.84%)and 65-85 years(23.64%).Reports were primarily originated from the United States(25.45%).Ticagrelor accounted for the highest number of reports(426 cases,45.37%).Among the nine antiplatelet drugs included,seven positive signals for in-stent thrombosis adverse events were generated,ticagrelor,tirofiban,and prasugrel demonstrated stronger risks of in-stent thrombosis,while aspirin showed a weaker risk.The detection results from the ROR and PRR methods were consistent.Conclusion Antiplatelet drugs may increase the risk of in-stent thrombosis.Aspirin is currently considered relatively safe.Incomplete inhibition of the purinoceptor 12 receptor is a risk factor influencing the development of in-stent thrombosis.关键词
