Huoxin Pills Alleviates Pathological Changes of Lung Tissue in Rats with Pulmonary Arterial Hypertension via Regulating Oxidative Stress

摘要

Abstract

Objective To observe the effects of Huoxin Pills(concentrated pills)on the pathological changes of lung tissue and oxidative stress in the rat pulmonary arterial hypertension model induced by monocrotaline(MCT).Methods Totally 48 male SD rats were randomly divided into control group(8 rats)and MCT group(40 rats),pulmonary arterial hypertension model was established in rats in the MCT group by a single intraperitoneal injection of MCT 60 mg/kg induced for 14 days.Totally 40 rats with successful modeling were randomly divided into the model group,the sildenafil group,and the low,medium,and high dose groups of Huoxin Pill,with 8 rats in each group.The sildenafil group was given sildenafil 25 mg/kg by gavage,and the low,middle and high dosage groups of Huoxin Pill were given Huoxin Pill(concentrated pills)solution of 5.4,10.8 and 21.6 mg/kg by gavage.The control group and model group were gavaged with the equal volume of distilled water once daily for 14 consecutive days.After intervention,the modified right heart catheterization was used to detect right ventricular systolic pressure(RVSP)and mean pulmonary arterial pressure(mPAP)in rats,and the right ventricular hypertrophy index(RVHI)was calculated for each group.The contents of superoxide dismutase(SOD)and malondialdehyde(MDA)in rat serum,as well as the activities of glutathione peroxidase(GSH-Px)and catalase(CAT)were detected by ELISA.HE staining was used to observe the pathological morphological changes of lung tissues in each group,Masson staining was applied to evaluate the degree of lung tissue fibrosis,and immunofluorescence was employed to detect the expression level of α-smooth muscle actin(α-SMA)in lung tissues for assessing the degree of pulmonary arteriole muscularization.Results Compared with the control group,rats in the model group exhibited pronounced cyanosis in the oronasal region and extremities.Pathological examination revealed swelling of the pulmonary arteriole walls,fibrous thickening of the intima,partial stenosis or even occlusion of the lumen,extensive infiltration of inflammatory cells,proliferation of blue collagen fibers around pulmonary arteries,increased collagen deposition area,and elevated expression of α-SMA protein,predominantly in fully muscularized vessels.These symptoms were alleviated to varying degrees in the intervention group.The model group showed significantly higher levels of RVSP,mPAP,RVHI,and serum MDA,while the activities of serum SOD,GSH-Px,and CAT markedly decreased(P<0.01)compared with the control group.When compared with the model group,the sildenafil group and the medium,and high dose groups of Huoxin Pill demonstrated significant reductions in RVSP,mPAP,RVHI,and serum MDA levels,accompanied by notable increases in SOD,GSH-Px,and CAT activities(P<0.05,P<0.01).Conclusion Huoxin Pill(concentrated pill)can reduce pulmonary artery pressure and ameliorate pathological damage of lung tissues in MCT-induced pulmonary arterial hypertension(PAH)model rats,and the mechanism may be related to the regulation of oxidative stress response.
