Effects of Cis-3-hexenal in Promoting Biosynthesis and Accumulation of Astragalosides in Astragalus membranaceus var.mongholicus Root Cultures

Abstract
Objective:To optimize the liquid culture system of Astragalus membranaceus var.mongholicus adventitious roots in the presence of cis-3-hexenal,establish a sample preparation method for measuring astragalosides in the culture solution and clarify the impact on the accumulation and release of astragalosides.Methods:Effects of the culture solution volume(20 and 30 mL),the inoculum size(5.0%,7.5%,and 10.0%of the culture solution volume),and the concentration of cis-3-hexenal(12.5,50.0,and 200.0 μmol·L-1)on the root growth and the content of astragalosides after four weeks of shake cultivation were examined to optimize the culture system.D101 macroporous resin was used for impurity removal,enrichment,and preparation of the culture solution samples.By using the optimized system,root cultures were processed by cis-3-hexenal for three days,and the contents of astragalosides in the root cultures and in the culture solution were dynamically determined by LC-MS/MS.Results:When the culture solution volume was 20 mL,and the inoculum size was 1.0 g,the application of 12.5 μmol·L-1 cis-3-hexenal promoted the root growth and the accumulation of astragalosides to the highest level.A column volume of 30 mL of D101 macroporous resin could completely adsorb astragalosides in 20-200 mL culture solution,and astragaloside Ⅳ,isoastragaloside Ⅰ,isoastragaloside Ⅱ,and isoastragaloside Ⅳ were mainly released into the culture solution.The treatment with 12.5 μmol·L-1 cis-3-hexenal for 4-8 h facilitated the most effective accumulation of astragaloside Ⅰ and cycloastragenoside Ⅱ.Conclusion:When the culture solution volume was 20 mL,adding 12.5 μmol·L-1 cis-3-hexenal to the liquid culture system of Astragalus membranaceus var.mongholicus adventitious roots is beneficial to the growth of the root cultures and the accumulation of astragalosides.关键词
