黄芪6个活性成分含量测定及与根系形态特征的相关性研究
Content Determination of Six Active Components in Astragali Radix and Its Correlation with Root Morphological Characteristics

Abstract
Objective:An ultra-high performance liquid chromatography-triple quadrupole mass spectrometry(UPLC-TQ-MS)method was employed for the simultaneous determination of six active components(astragaloside Ⅳ,calycosin,calycosin-7-O-β-D-glucoside,formononetin,ononin,and rutin)in Astragali Radix.Additionally,the correlation between the active components and root morphological characteristics of Astragali Radix was investigated.Methods:The effects of various elution gradients and flow rates on peak shape,resolution,and mass spectrometry response values were evaluated.The optimal conditions for gradient elution were established,with acetonitrile solution containing 0.1%formic acid at a flow rate of 0.4 mL·min-1.An ACQUITY UPLC BEH C18 column(100 mm×2.1 mm,1.7 μm)was employed,and mass spectrometry scanning was performed in electrospray ionization(ESI)mode.The contents of the active components in 48 samples of Astragali Radix were determined.The root length,root diameter,and root dry weight were measured,followed by correlation and stepwise regression analyses.Results:The six active components exhibited good linear relationships within their respective mass concentration ranges(r≥0.996).The relative standard deviation(RSD)values of precision ranged from 1.18%to 2.88%,and those of repeatability ranged from 1.42%to 3.05%.The average recovery rates were between 97.24%and 102.90%.The contents of astragaloside Ⅳ,calycosin,calycosin-7-O-β-D-glucoside,formononetin,ononin,and rutin were 0.576-2.729,0.033-0.496,0.054-0.762,0.006-0.080,0.066-0.846,0.000 1-0.000 3 mg·g-1,respectively.Correlation and stepwise regression analyses revealed that the contents of active components in Astragali Radix were associated with root morphological indices.Specifically,the content of astragaloside Ⅳ was significantly negatively correlated with the root diameter and root dry weight,while the content of rutin was significantly negatively correlated with the root dry weight.As root diameter increased,the content of astragaloside Ⅳ decreased,whereas the content of calycosin-7-O-β-D-glucoside increased.Conclusion:The UPLC-TQ-MS method established for the determination of active components demonstrates excellent linearity,repeatability,precision,and recovery rates,enabling the simultaneous quantification of the six active components in Astragali Radix.Furthermore,this research indicates that root morphological indices of Astragali Radix can serve as a reference for assessing the content of its active components.关键词
