Optimization of Extraction Process of Total Saponins from Astragalus membranaceus Fibrous Roots

Abstract

Objective:To optimize the ultrasonic-assisted extraction process of total saponins from Astragalus membranaceus fibrous roots using response surface methodology.Methods:The effects of ethanol volume fraction,solid-to-liquid ratio,extraction time,and extraction temperature on the extraction yield of total saponins from A.membranaceus fibrous roots were investigated through single-factor experiments.Based on the single-factor results,key factors affecting the extraction yield of total saponins from A.membranaceus fibrous roots were identified,and their optimal value ranges were determined.Using ethanol volume fraction,solid-to-liquid ratio,extraction time,and extraction temperature as independent variables,a four-factor,three-level optimization experiment was designed using response surface methodology.It was then employed to optimize the extraction process.Results:The single-factor experiments indicated that the optimal extraction conditions were an ethanol volume fraction of 60%,a solid-to-liquid ratio of 1:16,an ultrasonic extraction time of 25 min,and an extraction temperature of 60℃.Regression model analysis showed that the optimal extraction conditions for the extraction yield of total saponins from A.membranaceus fibrous roots were an ethanol volume fraction of 60%,a solid-to-liquid ratio of 1:16,an ultrasonic extraction time of 25 min,and an extraction temperature of 57℃.Under these conditions,the extraction yield of total saponins from A.membranaceus fibrous roots reached 2.82%.Conclusion:An efficient extraction technology for total saponins from A.membranaceus fibrous roots was established,providing a reference for the high-efficiency utilization and development of A.membranaceus resources.
