Quality Evaluation of Different Parts of Sarcandra glabra Based on Characteristic Chromatographic Profile by QAMS

Abstract
Objective:To establish a method for the quality evaluation of different parts of Sarcandra glabra.Methods:Chromatographic analysis was performed on an UPLC system equipped with a Waters BEH C18 column(100 mm×2.1 mm,1.7 μm)using a gradient elution of acetonitrile(A)and 0.1%aqueous phosphoric acid(B).The detection wavelength was set at 330 nm,and the column temperature was maintained at 25℃.Ultra-performance liquid chromatography(UPLC)characteristic chromatographic profiles of the aerial part,stem,and leaf of S.glabra were established,with a multi-components by single marker(QAMS)method for the neochlorogenic acid,chlorogenic acid,cryptochlorogenic acid,caffeic acid,isofraxidin,and rosmarinic acid.Similarity evaluations were performed,and orthogonal partial least squares discriminant analysis(OPLS-DA)was employed to assess and differentiate the chemical profiles of the different plant parts.Results:A total of 11 common peaks were identified in 42 batches of S.glabra from different parts,with similarity values all exceeding 0.840.OPLS-DA classified the different medicinal parts into three distinct groups,identifying three differential markers.There was no significant difference between the results obtained by QAMS and the external standard method.The contents of chlorogenic acid and isofraxidin in stems were higher than those in leaves,while the contents of neochlorogenic acid,cryptochlorogenic acid,caffeic acid,and rosmarinic acid in leaves were higher than those in stems.Conclusion:The established UPLC characteristic chromatographic profile and QAMS method can simultaneously evaluate different parts of S.glabra,providing a reference for composition analysis and quality control.关键词
