Digital Characterization of Trait Identification and Fluorescence Microscopic Features of Astragalus membranaceus var.mongholicus under Different Planting Patterns in Shanxi Province

Abstract
Objective:To investigate the macroscopic traits and microscopic characteristics of Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao under different cultivation patterns and establish an identification method based on digital characterization.Methods:The surface and cross-sectional structures of the samples were observed using stereo fluorescence microscopy,optical microscopy,and a full-field high-resolution fluorescence live-cell imaging system under visible light,red fluorescence(TexRE),green fluorescence(EGFP),and ultraviolet light(DAPI)modes.Digital analysis of the structural features was performed based on color and morphology.Result:Wild-simulated direct-seeded and transplanted A.membranaceus var.mongholicus exhibited differences in external morphology.Their cross-sectional digital profiles were also distinct.Furthermore,stained samples revealed clear differences in the degree of lignification and starch content when observed under optical microscopy.Conclusion:Through multiple comparative analyses,methods for differentiating A.membranaceus var.mongholicus from different planting patterns based on macroscopic traits and microscopic features were established,providing a scientific basis and methodological framework for further research on this species.关键词
