Abstract

Objective:To predict and analyze the quality markers(Q-markers)of Qinbei Zhike granules using combination of traditional Chinese medicine(TCM)serum pharmacochemistry and network pharmacology.Methods:Ultra-high-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)was used to analyze the plasma samples from rats administered Qinbei Zhike granules and from a control group.Network pharmacology was applied to identify constituents absorbed into blood,screen core components and targets,and analyze the mechanism of Qinbei Zhike granules in treating cough,thereby predicting potential Q-markers.Results:A total of 2 159 compounds were identified in the granule test solution,and 326 constituents absorbed into blood were detected in the serum.Then,network pharmacology analysis identified 27 core components,35 core targets,and 148 drug-disease co-targets.Gene Ontology(GO)functional enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses were performed on the co-targets using the DAVID database,leading to the identification of the main functional pathways involved in the prevention and treatment of cough,including cancer pathways,lipid and atherosclerosis pathways,and prostate cancer pathways.Conclusion:Comprehensive analysis identified the following components as meeting the five fundamental principles for Q-markers:apigenin,wogonin,baicalin,isoliquiritigenin,licochalcone A,oroxylin A,licochalcone B,norwogonin,and hyperoside.They can be the Q-markers for Qinbei Zhike granules,providing a reference for quality evaluation standards.
