Abstract

Objective:To establish a characteristic chromatogram for malva seed formula granules using ultra-high performance liquid chromatography(UPLC)and determine the mass fraction of caffeic acid.Methods:Waters UPLC® BEH Shield RP18 column(100 mm×2.1 mm,1.7 μm)was used,with acetonitrile and 0.1%phosphoric acid aqueous solution as mobile phase for gradient elution.Column temperature was 30℃;detection wavelength was 322 nm;flow rate was 0.30 mL·min-1;sample volume was 3 μL.Based on the above chromatographic conditions,a characteristic chromatogram of the malva seed formula granules was established.By using the chromatographic peak of caffeic acid as the reference(S)peak,the similarity between the characteristic chromatogram of the standard decoction of malva seed and the characteristic chromatogram of malva seed granules was evaluated.Moreover,a method for determining the content of caffeic acid in malva seed formula granules was established.Results:A total of eight characteristic peaks were identified in the malva seed formula granules'characteristic chromatogram,with peak 2 corresponding to caffeic acid.The similarity between the characteristic chromatogram of eight batches of malva seed formula granules and the control characteristic chromatogram was greater than 0.950.The mass fraction of caffeic acid in eight batches of malva seed formula granules ranged from 0.128 to 0.304 mg·g-1.Conclusion:The established characteristic chromatogram and content determination method for malva seed formula granules are simple,with good repeatability and high accuracy,and they provide reference for effectively controlling the quality of malva seed formula granules.
